摘要
目的探讨脐带源间充质干细胞(hUMSCs)在体内移植后对小鼠肝纤维化的干预影响。方法用硫代乙酰胺(TAA)建立小鼠肝纤维化模型,将hUMSC尾静脉移植后,比较移植前后3天、7天血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)水平变化,Masson染色考察肝脏胶原的聚集。结果 hUMSCs移植后3天和7天均可有效改善由TAA诱导造成的小鼠体重的减轻和肝重的增加,明显降低了血清中ALT和AST的活性,使肝细胞结构形态趋于正常,抑制了胶原的沉积。结论 hUMSCs移植可有效改善肝纤维化的发生。
Objective To explore the effect of human umbilical cord mesenchymal stem cells(hUMSCs) transplantation against mice liver fibrosis in vivo.Methods Administration with thioacetamide(TAA) to induce mice liver fibrosis,observing the changes of ALT and AST activities in serum and the accumulation of collagen in Masson staining after hUMSCs transplantation 3 days and 7 days.Results Both the transplatation of hUMSCs 3 days and 7 days could effective improve the mice body weight and decrease the liver weight against TAA induction,significantly reduce the activities of ALT and AST in serum,and also inhibit collagen accumulation to the normal.Conclusion Transplantation of hUMSCs could effectively ameliorate liver fibrosis.
