转hLYZ基因山羊繁殖、遗传及hLYZ基因的表达稳定性 被引量:1

摘要 为研究人溶菌酶(hLYZ)转基因山羊繁殖情况及外源基因在不同世代中的遗传与表达稳定性,通过常规育种方法对转hLYZ基因山羊进行繁殖,并采用PCR、Western-blot和ELISA方法对其后代个体进行整合与表达鉴定。经过3个繁殖周期,共获得转hLYZ基因山羊F1代9只,转基因整合率28.13%;F2代140只,转基因整合率45.31%;F3代22只,转基因整合率40.00%。hLYZ基因在G0、F1、F2代羊乳汁中稳定表达,3个世代平均表达量分别为3.46 g/L、2.28 g/L和2.22 g/L。研究结果表明转hLYZ基因山羊能正常繁殖,hLYZ基因能在山羊群体中稳定遗传和表达。 The inheritance and expression stability of human lysozyme (hLYZ) gene in each generation of transgenic goat were studied through conventional breeding. PCR, Western-blot and ELISA were employed to identify the integration and expression of exogenous gene in offsprings. After three breeding cycles, 9 hLYZ transgenic individuals in F1 genera- tion, 140 individuals in F2 generation and 22 individuals in F3 generation were obtained, with the integration rates of 28. 13% , 45.31% and 40.00% , respectively, hLYZ in goat milk of Go , F1 and F2 generations was stably expressed, and the average expression levels were 3.46 g/L, 2. 28 g/L and 2. 22 g/L, respectively. The results confirmed that hLYZ transgenie goats reproduced normally, and exogenous hLYZ gene could inherit and stably be expressed in goats.
