摘要
目的 研究乳腺癌、膀胱癌和肾癌组织中c myc基因扩增情况。方法 采用聚合酶链式反应 (polymerasechainreac tion ,PCR ) ,对 48例恶性肿瘤c myc基因扩增进行研究 ,通过激光密度扫描仪对琼脂糖凝胶EB染色底片进行积分光密度分析。 结果 正常组织无c myc扩增 ,c myc与n myc +l myc比值 95 %可信区间为 0 .0 86 9~ 0 .6 2 5 7,乳腺癌组织c myc基因扩增阳性率为2 5 .0 0 % ,膀胱癌组织的c myc基因扩增阳性率为 43 .75 % ,肾癌组织c myc基因扩增阳性率为 37.5 0 % ,48例肿瘤组织c myc基因扩增总阳性率为 2 6 .5 6 %。结论 癌组织c myc扩增率高于正常组织 ,在不同肿瘤组织中扩增程度不同 ,与细胞潜在的增殖能力呈正相关 ,对肿瘤的发生、发展及预后 ,特别是指导临床基因治疗提供有用的资料。
Objective To study the amplification of c-myc gene in different tumors.Methods c-myc gene were detected in 48 cases of malignant tumor by polymerase chain reaction(PCR),and the results in films staining with EB were analysised by integraling density.Results The amplification of c-myc gene was not found in 16 normal controls,and the positive rate of c-myc amplification was 25.00% in 16 breast cancers,43.75% in 16 breast cancers,37.50% in 16 renal cell carcinoma.Conclusion The results showed that c-myc gene is related with cell growth,differentiation and tumorous prognosis,the deteting c-myc amplification is useful for gene therapy with antisense oligonucleotide.
