低表达泛素连接酶CHIP对脑胶质瘤细胞U87迁移、侵袭及增殖的影响

摘要 目的探讨低表达泛素连接酶CHIP对脑胶质瘤细胞U87迁移、侵袭和增殖的影响。方法运用ControlsiRNA、CHIP siRNA分别转染脑胶质瘤细胞U87,Western blot检测CHIP蛋白表达情况,CCK-8细胞增殖实验检测CHIP siRNA对U87细胞增殖的影响,Transwell chamber细胞迁移、侵袭实验检测CHIP siRNA对U87细胞迁移、侵袭的影响。结果与Control siRNA组相比,CHIP siRNA组U87细胞CHIP蛋白表达量降低,CHIP siRNA组U87细胞增殖和迁移、侵袭数增加(P均<0.05)。结论 CHIP siRNA靶向干扰了泛素连接酶CHIP的表达,CHIP作为肿瘤抑癌因子抑制脑胶质瘤细胞U87的迁移、侵袭及增殖。 Objective To investigate the effect of low expression of the ubiquitin ligase CHIP on the migration, inva- sion and proliferation of glioma cells. Methods Control siRNA and CHIP siRNA were used to transfect U87 glioma cells respectively. The expression of CHIP in U87 glioma cells was detected by Western blot. The influence on the proliferation of U87 glioma cells was measured by CCK-8 assay. The influences on the migration and invasion of U87glioma cells were measured by Transwell chamber cell migration and invasion assay. Results Compared with Control siRNA group, CHIP siRNA group could reduce the expression level of CHIP in U87glioma cells, and the number of cell migration, invasion and proliferation in U87 glioma cells was increased. Conclusions CHIP siRNA targeting interferes the expression of the ubiq- uitin ligase CHIP. As a tumor suppressor factor, CHIP suppresses the migration, invasion and proliferation of U87 glioma cells.
