嗜水气单胞菌Ⅲ型分泌系统AscV基因的检测与序列分析 被引量:3

Detection and Sequence Analysis of AscV Gene of Type Ⅲ Secretion System(TTSS) in Aeromonas hydrophila Strains

摘要 对分离于草鱼、青鱼、鲤鱼、牙鲆、宽体金线蛭、中国林蛙的致病性嗜水气单胞菌(Aeromonas hy-drophila),进行Ⅲ型分泌系统(typeⅢsecretion system,TTSS)AscV基因的检测与分析,探讨不同来源嗜水气单胞菌Ⅲ型分泌系统AscV基因的携带与分布情况。利用文献发表的TTSS的AscV基因特异性引物,采用PCR方法扩增该基因片段,选取代表菌株的AscV基因序列,以DNA Star软件与GenBank公布的参考菌株相应基因序列进行同源性比较,构建系统进化树。结果在供试的42株菌中,有34株携带(阳性率80.95%);其中分离于宽体金线蛭的10株有8株携带(阳性率80%)、中国林蛙的11株有10株携带(阳性率90.9%)、牙鲆的10株有9株携带(阳性率90%)、草鱼的7株有3株携带(阳性率42.9%)、青鱼和鲤鱼的各2株均携带(阳性率100%)。所检测的5个代表菌株AscV基因与参考菌株的同源性在81.0%~98.7%之间;供试不同来源菌株之间的同源性在91.2%~99.3%之间。 In this study,the pathogenicity of Aeromonas hydrophila strains isolated from Ctenopharyngodon idellua,Mylopharyngodon piceus,Cyprinus carpio,Paralichthys olivaceus,Whitmania pigra and Rana temporaria chensinensis were investigated for the presence of AscV gene of the type III secretion system(TTSS),and the carrying and distribution of AscV of Aeromonas hydrophila isolated from different hosts were discussed.Using specific primers published in the reference,the gene fragments of AscV of above strains were obtained by PCR methods,the homology of AscV gene from diverse representative A.hydrophila strains were compared with available sequences in NCBI database and phylogenetic tree was constructed using DNAstar software.The results showed 34 in all 42 detective strains carried AscV gene,the positive rate was 80.95% including 8 in all 10 strains isolated from Whitmania pigra(positive rate 80%),10 in all 11 strains isolated from Rana temporaria chensinensis(positive rate 90.9%),9 in all 10 strains isolated from Paralichthys olivaceus(positive rate 90%),3 in all 7 strains isolated from Ctenopharyngodon idellua(positive rate 42.9%),2 in all 2 strains isolated from Mylopharyngodon piceus(positive rate 100%) and 2 in all 2 strains isolated from Cyprinus carpio(positive rate 100%).The nucleotide homologies between the AscV gene of five representative A.hydrophila strains and that of the reference strains was 81.0%-98.7% and the nucleotide homologies among A.hydrophila strains isolated from different hosts were 91.2%-99.3%.
