摘要
目的观察桂枝茯苓胶囊(桂枝、茯苓、牡丹皮、芍药、桃仁)对动脉粥样硬化大鼠心脏基质金属蛋白酶-2(MMP-2)表达和微小血管密度的影响。方法 SD大鼠随机分为空白组(12只)和造模组(38只)。空白组予普通饲料;造模组予高脂饲料+维生素D3腹腔注射,造模成功后造模组随机分为模型组、西药对照组、治疗组。分别予生理盐水、辛伐他汀合卡托普利、桂枝茯苓胶囊灌胃,14周末处死动物。检测心脏微小动脉和毛细血管密度、MMP-2。结果模型组心肌微小动脉密度、MMP-2表达明显高于空白组,毛细血管密度明显低于空白组(P<0.05);与模型组比较,治疗组与西药对照组微小动脉密度、MMP-2表达明显降低,毛细血管密度明显增高(P<0.05),两组比较,微小动脉密度、毛细血管密度及MMP-2表达无显著性差异(P>0.05);MMP-2表达与微小动脉密度呈显著正相关,与毛细血管密度呈显著负相关(P<0.05)。结论桂枝茯苓胶囊能改善动脉粥样硬化大鼠心脏微小血管病变,机制可能与抑制MMP-2表达有关。
AIM To observe the effect of Guizhi Fuling Capsules(Cinnamomi Ramulus,Poria,Moutan Cortex,Paeoniae Radix,Persicae Semen) on expressions of matrix metalloproteinase-2(MMP-2) and change of microvascular density in rats with atherosclerosis.METHODS Fifty healthy SD male rats were randomly divided into two groups,normal group and marking model group.Rats in the normal group were fed with ordinary diet.Rats in marking model group were established by high cholesterol diet and intraperitoneal injection of Vitamin D3,and were randomly divided into three groups,atherosclerosis group,control group and therapy group,which were given 0.9% NaCl,simvastatin plus captopril and Guizhi Fuling Capsules by intragastric administration,respectively.The rats were killed at the 14th weekend.Immunohistochemistry was used for observing the expression of MMP-2,arteriole and capillary in myocardium.RESULTS The arteriole density and the amount of MMP-2 expression in atherosclerosis group were higher than those in the normal group(P〈0.05).The capillary density in atherosclerosis group was lower than that in the normal group(P〈0.05).Compared with atherosclerosis group,the arteriole density and the amount of MMP-2 expression significantly reduced and the capillary density significantly increased in both treatment group and control group(P〈0.05).There was no significant difference between the two groups(P〉0.05).MMP-2 expression was positively correlated with the arteriole density and was negatively correlated with the capillary density(P〈0.05).CONCLUSION Guizhi Fuling Capsules can improve the cardiac microvascular lesion in rats with atherosclerosis.The mechanism may be related to inhibiting the expression of MMP-2.
