摘要
目的研究阿魏酸钠(sodium ferulate,SF)能否对抗脂多糖(lipopolysaccharide,LPS)所致的大鼠海马炎症反应。方法 SD大鼠分为对照组、LPS组、SF组,SF组每天灌胃给予SF(100、200 mg.kg-1),对照组和LPS组灌胃等量生理盐水,3 w后,脑室注射LPS建立损伤模型,对照组注射等量生理盐水。脑室注射7d后,制备大鼠脑石蜡切片,进行Nissl染色和胶质纤维酸蛋白的免疫组织化学染色,取海马CA1区,Western蛋白印迹观察IL-1β和Caspase-3表达。结果与LPS组相比,SF(100、200 mg.kg-1)能减轻海马锥体神经元的损伤,抑制星形胶质细胞激活及IL-1β、Caspase-3的表达(P<0.01)。结论 SF能通过抑制LPS引起的IL-1β表达,减轻海马炎症反应及神经元损伤。
Objective To study whether the sodium ferulate （SF） can fight LPS - induced inflammatory reaction in rat hippocampus. Methods SD Rats were divided into the control group, LPS group and SF group. The SF group were injected with SF （100, 200 mg· kg-1 ） every day, the control group and LPS group were given the same volume of saline. Three weeks later, ventricles were injected with LPS to establish damage model, and the control group were injected with saline of the same volume. Seven days after the injection, paraffin section of rat brain was made to observe the morphological change in hippocampal CA! region by means of Nissl staining and immunocytochemical staining. Glial fibrillary acidic protein （GFAP） were used to determine the astrocyte activation. Western blotting was used to determine the expression of IL- 1β, Caspase -3 in hippocampal CA1 region. Results Compared with LPS group, SF （100, 200 mg·kg-1） could relieve the injury in hippocampal pyramidal neuron and inhibit the expression of astrocyte activation and IL - 1β, Caspase- 3 （P 〈 0. 01 ）. Conclusion SF can suppress inflammatory reaction in hippocampus and relieve the neuron damage by inhibiting the expression of LPS - induced IL - 1β.
