摘要
目的探讨骨组织提取液谷慷太林对大鼠骨折愈合的影响、垂体功能的调节作用及其促进骨折愈合的机制。方法取54只健康成年雌性SD大鼠(200±20)g,6只为空白对照组(B组),其余48只用定点手法折骨术造成大鼠双侧桡骨中段闭合性骨折,随机分为谷慷太林治疗组(G组)和骨折模型对照组(M组),每组24只。分别于第7、14、28、42天后各组随机处死6只大鼠,运用X线观察骨折愈合情况,利用骨密度测量仪检测骨折端骨密度值(BMD),并采用HE染色光镜下观察骨折处骨痂,运用免疫组化技术观察大鼠垂体中促甲状腺激素(TSH)和生长激素(GH)的阳性细胞数。结果 X线平片检查显示第14、28、42天时G组骨痂形成较M组明显;BMD结果显示骨密度值在各时间段G组均比M组高(P<0.05);骨痂组织学检查显示G组相对于M组断端周围的骨小梁数量增多,宽度较大,排列整齐,其形成的骨痂成熟。大鼠垂体中GH与TSH阳性细胞数的峰值在时间上G组均早于M组出现,尤其以第14、28天为明显(P<0.01)。结论骨组织提取液谷慷太林注射液具有促进骨折愈合的作用,其促进骨折愈合的途径之一可能与促进垂体合成与分泌生长激素、促甲状腺激素有关。
Objective To explore the effects of soluble muhi-peptide agent, marked Gu Kang Tai Lin,derived from bone tissue on rat bone fracture healing at different time intervals. To observe the functional changes of adjustment in pituitary and discuss the mechanisms of promoting union of fracture with Gu Kang Tai Lin. Methods Fifty four female adult SD rats (200±20, weight g) were used for this investigation. Closed fracture of radius of the rest forty eight female rats were produced except for six rats in the blank group. Then they were divided randomly into 2 groups: the GuKangTaiLin group and the Model group. Six rats in each group were executed on the 7th, 14th, 28th and 42nd day after operation to observe the radiographic findings ; bone mineral density (BMD) measurements ; histological and morphological chan ges in osteotylus; the TSH cells and GH cells in pituitaries were showed by immunocytochemical method. Results At different time intervals the radiographic findings, bone mineral density (BMD) measurements of the fractural indicated that the Gu Kang Tai Lin group had more bony calluses than the Model group (P d0.05) ; And histological changes demonstrated that bone trabecula of the Gu Kang Tai Lin group were more mature than those in the model group. At broken ends of fractured bone, a great deal of bone trabec- ula around the bone callus line up in order. On the other hand, the results of statistical analysis showed: The peak amplitude of GH cells and TSH cells population of the Gu Kang Tai Lin group were obviously anterior to the model group on the 14th and 28th day (P 〈0.05). Conclusion The bone tissue extract Gu Kang Tai Lin injection strongly accelerate rat fracture healing and improve the quality of healing. The mechanism of Gu Kang Tai Lin injection to advance fracture healing were possibly concerned with control of hypophysis through synthesizing and secreting growth hormone and thyroid stimulating hormone.
