摘要
目的:研究重组人肿瘤坏死因子受体融合蛋白(rhTNFR:Fc)对佐剂性关节炎(adjuvant arthri-tis,AA)大鼠滑膜细胞增殖和分泌功能的影响。方法:完全弗氏佐剂诱导SD大鼠AA模型,3H-TdR掺入法检测滑膜细胞增殖,ELISA法测定滑膜细胞上清中白细胞介素1β(IL-1β),IL-6,IL-17,淋巴细胞毒素(LTα),NF-κB配体的受体或激活因子(RANKL),基质金属蛋白酶-13(MMP-13)水平。结果:rhTNFR:Fc体外给药可以抑制TNF诱导的滑膜细胞增殖,下调IL-1β,IL-6,IL-17,LTα,RANKL,MMP-13的分泌水平。结论:rhTNFR:Fc抑制滑膜细胞增殖和分泌功能可能是其治疗AA的作用机制之一。
Objective: To investigate the effect of rhTNFR: Fc on proliferation and cytokines production of synoviocytes from adjuvant arthritis (AA) rats. Methods: AA rats were induced by complete Freund adjuvant. The proliferation of synoviocytes was detected by 3H-TdR. The levels of IL-113, IL-6, IL-17, LTα, RANKL and MMP-13 in supernate of synoviocytes were assessed by ELISA. Results: Administration of rhTNFR: Fc significantly inhibited the proliferation of synoviocytes and decreased the secretion of IL-lβ, IL-6, IL-17, LTα, RANKL and MMP-13. Conclusions: Our results indicate that rhTNFR:Fc significantly reduces the proliferation and proinflamma- tory cytokines production of synoviocytes, which might be one of the therapeutic mechanisms for rhTNFR:Fc in AA.
