摘要
采用KMnO4作为固定剂,对传统的免疫荧光细胞化学技术进行改进,将制备好的样品用激光扫描共聚焦显微镜进行连续断层扫描和三维重建,清晰地呈现了8种纤毛虫的表膜以及纤毛模式,为原生动物纤毛虫表膜研究提供了一种可供借鉴的样品制备方法。
By using KMnO4 as fixative,the traditional immune-fluorescence cytochemistry technology can be improved.The prepared sample was studied by continue section scanning and three dimensional reconstruction with confocal scanning laser microscopy.The eight ciliate membranes and the Ciliate mode can be clearly observed.This method provides a reference for the sample preparation of protozoon ciliate membranes.
