青格里绒山羊微卫星标记遗传多态性及其与部分经济性状相关关系的研究 被引量:1

摘要 【目的】研究青格里绒山羊群体的遗传多态性,检测并分析多态标记与生产性状的关联性,找到与生产性状相关的遗传标记,为标记辅助选择育种提供理论依据。【方法】采用微卫星标记技术研究9个微卫星标记在青格里绒山羊群体中的遗传多态性,利用Popgene(Version1.32)软件计算等位基因频率(P)、基因杂合度(H)、有效等位基因数(Ne);用PIC-CALC软件计算多态信息含量(PIC),并用SAS软件的一般线性模型(GLM)分析多态位点与部分生产性状的相关性关系。【结果】9个微卫星标记中,微卫星标记BM6438没有多态性,其它8个微卫星标记均存在多态性。在青格里绒山羊群体中,8个有多态性的微卫星标记的平均有效等位基因数(Ne)、杂合度(H)、纯合度(P)和多态信息含量(PIC)分别为4.821 9、0.784 9、0.215 1、0.754 0。关联分析表明:绒细度性状上,标记OarHH35的BF基因型个体的绒细度显著低于其它基因型个体(P<0.05),标记BM3033的DF基因型个体极显著低于CE基因型个体(P<0.01),标记BM1824的BE基因型个体极显著低于AB基因型个体(P<0.01);产绒量性状上,标记BMS1788的CG基因型个体显著高于BC和EG基因型个体(P<0.05);标记BM1824的AD和BE基因型个体显著高于CF基因型个体(P<0.05)。【结论】与生产性状相关联的4个微卫星标记OarHH35、BM3033、BM1824、BMS1788可望作为青格里绒山羊绒细度或产绒量的分子遗传标记。其中,标记BM1824的BE基因型可望作为同时影响青格里绒山羊绒细度和产绒量的分子标记,这对于绒山羊分子育种研究工作的开展具有重要意义。 [ Objective ] To study the genetic polymorphism about nine microsatellite markers, and the correlation between genetic marker and production traits was analyzed by using general linear model (GLM) in Qinggeli cashmere goat population. This study was designed to provide certain theoretical basis for marker assisted selection (MAS). [ Method]The genetic diversity of nine microsatellite markers was studied by using microsatellite marker technology in Qinggeli cashmere goat population. Gene frequency (P), gene heterozygosity (H) and effective number of alleles (Ne) were calculated by Popgene software (release 1.32) ,and polymorphism information content (PIC) was calculated by PIC - CALC software. Correlation between genetic marker and production traits were analyzed by means of general linear model (GLM) [ Result ] The resuhsshowed that, among the nine microsatellite markers, loci microsatellite BM6438 was non - polymorphism loci in Qinggeli cashmere goats, the average effective numbers of alleles (Ne), gene heterozygosity ( H), Gene homozygosity ( Ho ), polymorphism information content ( PIC ) respectively were 4. 821 9, 0.784 9, 0. 215 1, 0.754 0, respectively. The Correlation analysis results showed that as for the fineness genotype BF of OarHH35 was significantly lower than those of other genotypes ( P 〈 0.05 ), genotype DF of BM3033 was extremely significantly lower than that of CE genotype ( P 〈 0.01 ) , genotype BE of BM1824 was extremely significantly lower than that of AB genotype(P 〈 0.01 ) ; as for the yield, genotype CG of BMS1788 was significantly higher than that of BC and EG genotypes(P 〈0.05) ; genotypes AD and BE of BM1824 were significantly higher than those of CF genotype( P 〈 0.05 ). [ Conclusion] Four microsatellite loci OarHH35, BM3033, BM1824, BMS1788 which all have correlation with production traits can be used to assess goat genetic diversity and regarded as genetic marker of cashmere fineness and yield choice in th
