摘要
【目的】研究4个微卫星标记在3个绵羊群体中的遗传多态性,以期找到与生长性状相关的遗传标记,为标记辅助选择育种提供理论依据。【方法】采用微卫星标记技术研究4个微卫星标记在3个绵羊群体(中国美利奴羊、中国美利奴羊与德国美利奴羊的杂交后代、中国美利奴羊与萨福克羊的杂交后代)中的遗传多态性,利用Popgene(Version1.32)软件计算等位基因频率(P)、基因杂合度(H)、有效等位基因数(Ne);用PIC-CALC软件计算多态信息含量(PIC)。【结果】4个微卫星标记在3个绵羊群体中共检测到26个等位基因,平均每个标记检测到6.5个等位基因,平均有效等位基因数(Ne)分别为4.384 8、5.452 0、5.006 8,平均杂合度(H)分别为0.771 3、0.812 2、0.799 6,平均多态信息含量(PIC)分别为0.742 2、0.785 3、0.770 6。【结论】4个微卫星标记在3个绵羊群体中均表现为高度多态,可以用作绵羊遗传多样性的评估,可望作为生长性状选择的遗传标记。
[ Objective] To research on the genetic polymorphism about four microsatellite markers in three sheep population in order to find out genetic marker associated with growth traits and provide certain theory basis for marker assisted breeding. [ Method ] In the three sheep population (Chinese Merino, crossbred population of a Chinese Merino and a German Merino, crossbred population of a Chinese Merino and a Suffolk), the genetic diversity of four microsatellite markers is studied by using microsatellite marker technology. Gene frequencies ( P), gene heterozygosity ( H), effective number of alleles (Ne) are calculated by Popgene software (Versionl. 32 ), and polymorphism information content (PIC) is calculated by PIC - CALC software. [Result]The results showed that in the three sheep population, there are 26 alleles in four microsatellite markers,and on average there are 6.5 alleles for every microsatellite marker. Average effectivenumber of alleles ( Ne ) respectively is 4. 384 8, 5. 452 0, 5. 006 8 ; average gene heterozygosity ( H ) respectively is 0. 771 3, 0.812 2, 0. 799 6; average polymorphism information content(PIC) respectively is 0.742 2, 0. 785 3, 0. 770 6. [ Conclusion] It is shown that the four microsatellite markers belong to high polymorphism in the three sheep population belong on high polymorphism and they can be used as an assessment with sheep' s genetic diversity and regarded as ~enetic marker of ~rowthtraits chnice.
