摘要
【目的】通过观察芜菁花蕾大小与花粉发育时期的对应关系,了解花粉发育动态,以提高单倍体育种取材的准确性。【方法】DAPI荧光染色后通过荧光显微镜镜检,观察花粉发育情况。【结果】供试品种花蕾长1.0~1.99 mm时花粉细胞大部分处于四分体时期,个别处于单核早期,单核靠边期的比例为0;蕾长2.0~2.49 mm时,花粉细胞随着内容物的增多和液泡的形成变大,细胞核被挤到细胞边缘,靠近细胞壁,单核靠边期比例较高;蕾长2.5~2.99 mm时,花粉细胞继续膨大,细胞正圆形,细胞核进行有丝分裂产生一个较小的生殖核和一个较大的营养核,细胞进入双核期;蕾长3.0~3.99 mm、4.0~4.99 mm时,花粉细胞继续增大,三核期居多。【结论】当花蕾长度在2.0~2.49 mm,单核靠边期比例最高,为62.40%。
[ Objective ] The studies on relationship between size of flower and pollen development in turnips,with the purpose of providing a way to know the pollen development dynamic, and improving the accuracy of haploid breeding. [ Method] DAPI fluorescent dye, through the fluorescence microscope test, to observe pollen development states. [ Result ] when the bud length of the turnip is in 1.0 - 1.99 mm, the pollens are mostly in tetrad period, the proportion of Uninucleate microspores at periphery is in 2.0 - 2.49 mm, the pollen was filled out with the content and, the nucleus was crowded to the cell edge, close to the cell wall, the ratio of Uninucleate microspores at periphery is 62.52% ; in 2.5 -2.99 mm, pollen cells continue to swollen, cells are become round, nuclei are mitotic, and produce a small generative nucleus and a large vegetative nucleus, cells into dual - core period ; when in 3.0 - 3.99 mm, 4.0 - 4.99 mm, pollen cells continue to expand,trinucleate stage is the majority. [ Conclusion]When the flower bud length is in 2.0 -2. 49 mm, the proportion of Uninueleate microspores at periphery is 62.52%.
