摘要
目的探索抗 TCRαβ单抗诱导异基因成年小鼠皮肤移植耐受的作用。方法经 C5 7BL / 6 (H - 2 b,B6 )小鼠尾静脉注射 BAL B/ c小鼠 (H - 2 d)脾细胞 ,2 d后腹腔注射环磷酰胺 (CP) ,随后两次尾静脉注射抗小鼠 - TCRαβ的单克隆抗体 ,然后进行皮肤移植 ,观察皮肤存活期并对耐受小鼠的 ML R、DTH等耐受状态进行了检查。结果耐受 B6小鼠获得了供体特异性皮肤移植耐受 ,供体皮肤移植存活时间明显延长 ,ML R和 DTH检查证明 B6小鼠对 BAL B/ c小鼠的脾细胞的反应性、特异性降低 ,而仍保持对无关第三者小鼠的免疫反应。结论使用注射脾细胞 ,环磷酰胺及抗 TCRαβ单抗的方法在
ObjectiveTo explore the method of inducing the tolerance of skin grafts between allogeneic adult mice. MethodsThe spleen cells of BALB/c mice (H 2 d) were injected to C57BL/6 mice (H 2 b,B6) via the tail vein. Two days later, an intraperioneal injection of cyclophosphamide (CP) was given to the B6 mice,and following two consecutive anti TCRαβ monoclonal antibody injections via the tail vein,skin grafts of BALB/c mice were transplanted to the B6 mice. To observe the survival time of the skin grafts,MLR and DTH were examined to confirm the tolerant state of B6 mice. ResultsThe tolerant B6 mice acquired specific tolerance to the skin grafts of the donor BALB/c mice. The survival time of the allogeneic skin grafts prolonged significantly. The results of MLR and DTH indicated that the B6 mice acquired specific tolerance to the spleen cells of the BALB/c mice,but the immune responses to the spleen cells of the irrelevent KM mice remained normal. ConclusionThe spleen cells,CP combined with anti TCRαβ monoclonal antibody can successfully induce the specific tolerance to the skin grafts of the BALB/c mice in the B6 mice.
