摘要
选用纤维素酶提取海带多糖并对提取多糖的抗氧化性进行了测试。通过单因素实验和响应面分析确定纤维素酶酶解提取海带多糖的最佳条件为:料液比1:40、纤维素酶添加量1.5%、温度44℃、pH值4.0、反应时间2h,在此条件下,海带多糖得率为(11.62±0.03)%。此外,ABTS自由基清除和铁氰化钾还原的抗氧化实验研究表明,乙醇沉淀得到的海带多糖具有一定的抗氧化活性,并且高浓度乙醇沉淀得到的褐藻淀粉抗氧化能力大于低浓度乙醇沉淀得到的褐藻糖胶和褐藻胶。
Cellulase-catalyzed hydrolysis was used to extract polysaccharides from Laminaria japonica Aresch and the antioxidant activities of the extracted polysaccharides were studied.The effects of cellulase concentration,the ratio of material to water,temperature,pH and time on the yield of polysaccharides were investigated by single factor experiments and response surface analysis.The optimal condition for the extraction was treating the material (a material-to-water ratio of 1:40) with 1.5% cellulase at 44 ℃ and pH4.0 for 2 h.Under this condition,the maximum yield of polysaccharides was obtained as (11.62±0.03)%.Moreover,the crude polysaccharides precipitated with ethanol demonstrated some antioxidant activity in the assays of ABTS radical scavenging activity and potassium ferricyanide reducing power.The result showed that the antioxidant capacity of the laminaran was greater than the fucoidan and alginate.
