摘要
对东北矮紫杉进行了愈伤组织的诱导,并建立了细胞悬浮培养系。研究表明,茎、叶均可诱导出愈伤组织,愈伤组织诱导培养基以MS+2,4-D 1.0 mg/L+NAA 1.0 mg/L+KT 0.1 mg/L为佳,愈伤组织继代培养基以B5+2,4-D 2.0 mg/L+KT 0.1 mg/L为佳。以嫩枝为外植体,愈伤组织的诱导率较高、状态较好,且较适于悬浮培养。悬浮培养的接种量2.0 g/瓶为最佳密度,细胞生长周期为20 d,对数生长期为10~16 d,细胞收获期为16d。试验中还发现,聚乙烯吡咯烷酮是东北矮紫杉组培中解除培养物褐化的较好抗氧化剂,使用浓度为1mg/L。
Callus were induced from T.cuspidata sieb.et Zucc.var. and suspension culture cell lines in vitro were set up. It indicated that callus could be induced through young shoot and needles.The rate of callus inducement in medium MS plus 1.0mg/L 2,4-D, 1.0 mg/L NAA and 0.1 mg/L KT were higher than others.The subculture of callus in medium B5 plus 2.0mg/L 2,4-D and 0.1mg/L KT were better than others. Induction frequency of callus from young shoot is higher than from needles. The callus from young shoot are better than from needles. The quantity of suspension culture cell is 2.0g/flask(25ml/flask). The cells growth cycle is 20 days, logarithmic growth phase being 10-16 days and harvest time being 16 days. It was observed that the polyvidone was an effective antioxidant removing brown of explants in tissue culture and the concentration is 1mg/L.
