摘要
目的探讨乳腺癌转移抑制基因(BRMS1)和运动相关蛋白(fascin)基因在肝癌组织中的表达及其临床意义。方法应用半定量逆转录.聚合酶链反应(RT—PCR)技术检测术中所取得的81例肝癌标本及其配对癌旁1.0cm肝组织和40例正常肝组织中BRMS1、fascin的mRNA表达。应用Westernblot法验证81例肝癌组织、配对癌旁组织和40例正常肝组织中BRMS1、fascin在蛋白水平的表达。结果肝癌组织中BRMS1mRNA的相对表达水平为0.4553±0.0451,低于正常肝组织的0.6124±0.0231和癌旁肝组织的0.6013±0.0175(P〈0.05);肝癌组织中fascinmRNA的相对表达水平为0.5047±0.0525,高于正常肝组织的0.4052±0.0152、癌旁肝组织的0.4130±0.0221(P〈0.05);2个基因在非癌组织中的表达水平差异均无统计学意义(P〉0.05)。BRMS1和fascin的蛋白表达定性实验表明:在肝癌组织和非癌肝组织中均有2种蛋白的表达,且BRMS1在肝癌组织中的表达低于非癌肝组织,而fascin在肝癌组织中的表达高于非癌肝组织。BRMS1和fascin基因的表达水平均与患者性别、年龄、肿块大小、病理类型无关(P〉0.05),而与淋巴结有无转移、临床分期相关(P〈0.05)。fascin/BRMS1的相对吸光度(A)值比值随肿瘤转移程度增高。结论在肝癌组织和非癌组织中BRMS1和fascin的mRNA和蛋白的表达是一致的。两基因的表达改变可能与肝癌侵袭转移密切相关;fascirr/BRMS1的相对A值比值可成为评估肝癌转移程度的有价值指标之一。
Objective To detect the expressions of breast cancer metastasis suppressor 1 ( BRMS1 ) and fascin ( an action-bounding protein) mRNA in the tissue of liver carcinoma, as well as to learn their clinical significance. Methods The expression of BRMS1 mRNA and fascin mRNA were detected by semi-quantitative reverse transeription-polymerase chain reaction (RT-PCR) in 81 cases hepatoma tissues and paired adjacent liver tissues to carcinoma and 40 cases normal hepatic tissues. The expression of BRMS1 and fascin in the level of protein in 81 cases hepatoma and paired adjacent liver tissues to carcinoma and normal hepatic tissues were detected qualitatively by Western blotting. Results The expression of BRMS1 mRNA in hepatoma tissue (0. 4553 ± 0. 0451 ) was lower than normal hepatic tissue and adjacent hepatic tissue to carcinoma (0. 6124 ±0. 0231 and 0. 6013 ±0. 0175) ; The expression of fascin mRNA in hepatoma tissue (0. 5047 ± 0. 0525 )was higher than normal hepatic tissue and adjacent hepatic tissue to carcinoma (0. 4052 ± 0.0152 and 0.4130 ± 0221 ). Qualitative experiments show that in liver cancer tissue and the non cancerous liver tissues are both exist the two kinds of protein expression, and BRMS1 expression in liver cancer tissue is lower than the non cancerous liver tissue, and fasein expression in liver cancer tissue was higher than non cancerous liver tissue. The expressions of BRMSI and fascin were neither correlated with sex, age, tumor size and pathological type; but was related with clinical stage and lymph nodes metastasis. The relative optic density ratio of fascin to BRMS1 increased with the development of metastasis. Conclusion BRMS1 and fascin mRNA and their protein' express were accordant; the expressions of BRMS1 and fascin are related to the Invasion and metastasis of liver carcinoma; The relative optic density ratio of fascin to BRMS1 may become an index for metastasis in liver carcinoma.
