三阴性乳腺癌组织DJ-1和PTEN及AR表达与预后相关性分析 被引量:15

Expression of DJ-1,PTEN and AR in triple-negative breast cancer and its correlation with relative clinical parameters and prognosis

摘要 目的:探讨三阴性乳腺癌(TNBC)组织中帕金森相关蛋白(DJ-1)、第10号染色体同源缺失性磷酸酶-张力蛋白基因(PTEN)和雄激素受体(AR)的表达及其与临床病理因素和预后的关系。方法:免疫组织化学SP法检测DJ-1、PTEN和AR蛋白在68例TNBC组织和30例正常乳腺组织中的表达,并分析其与TNBC临床病理因素的关系,及其对预后的影响。结果:TNBC组织中DJ-1阳性表达率为70.6%(48/68),AR阳性表达率为36.8%(25/68)。正常乳腺组织中DJ-1阳性表达率为6.7%(2/30),无AR表达。TNBC组织中DJ-1、AR阳性表达率明显高于正常乳腺组织,χ2值分别为7.09和6.72,P值分别为0.002和0.005。TNBC组织中PTEN阳性表达率为27.9%(19/68),正常乳腺组织中为100.0%。TNBC组织中PTEN阳性表达率显著低于正常乳腺组织,χ2=5.94,P=0.008。DJ-1、PTEN和AR表达在TNBC组织中的表达与腋窝淋巴结转移、组织学分化和TNM分期密切相关,P<0.01。TNBC组织中DJ-1和AR的表达均与PTEN蛋白的表达呈负相关,r=-0.529,P=0.009;而两者之间呈正相关,r=0.496,P=0.011。在5年无瘤生存率和5年总生存率方面,DJ-1和AR表达阴性者明显高于阳性者,P<0.01;PTEN表达阳性者高于阴性者,P<0.01。Cox回归多因素分析结果表明,腋窝淋巴结转移、DJ-1、PTEN及AR表达是影响TNBC患者5年无瘤生存率的独立预后因素,P<0.05。结论:TNBC组织中DJ-1和AR过表达,而PTEN表达下调,三者均为TNBC患者预后的独立危险因素,参与调控TNBC的发生与预后。 OBJECTIVE:To investigate the association among the expression of DJ-1,PTEN and AR in triple-nega- tive breast cancer tissues,and explore the correlation between the expression and its relative clinical parameters. METH- ODS: We collected 68 cases of TNBC and 30 cases of normal people to determin the expression of DJ-1 ,PTEN and AR by using immunohistochemical staining. RESULTS.. The positive expression rates of DJ-1 and AR in TNBC were 70. 6% (48/68) and 36.8% (25/68) respectively. In contrast,the positive expression of DJ-1 in normal tissues was only 6.7% (2/30). In normal tissues no AR expressed. Compare to normal tissues, the positive expression rates of DJ-1 and AR were significantly increased (Xz were 7.09 and 6.72,P were 0. 002 and 0. 005). The positive expression rate of PTEN in TNBC was only 27. 9% (19/68), but positive expression in normal tissues was 100. 0%. Compare to normal tissues, PTEN positive expression in TNBC was significantly decreased (X2 = 5.94, P = 0. 008). The expression of DJ-1, PTEN and AR in TNBC were significantly associated with lymph node metastasis,differentiation and TMN stages (P〈0.01). In 68 TNBC,DJ-1 and AR expression was negatively correlated with PTEN expressions (r= -0. 529, P= 0. 009). DJ-1was positively correlated with AR expression (r=0. 496 ,P=0. 011). Using Cox regression,lymph node metastasis,DJ-1, PTEN and AR were strong independent prognostic factors for five years of disease-free survival rate of TNBC patients (P〈0.05). CONCLUSIONS: DJ-1 ,PTEN and AR may contribute to the development of TNBC and aie valuable prognos- tic factors for TNBC patients. DJ-1 may regulate AR expression via inhibiting PTEN expression. The cooperation of these three proteins may be related to the prognosis of TNBC.
