摘要
目的应用17β-雌二醇(E2)建立雌性大鼠真性性早熟模型。方法 15日龄雌性SD仔大鼠采用ig给予雌二醇50μg.kg-1连续5 d制备性早熟模型。观察大鼠阴门开启平均日龄(VO)、第一个发情间期出现的平均日龄(D1)和性周期稳定后第一个发情期出现的平均日龄(E1);测定血清中黄体生成素(LH)、卵泡刺激素(FSH)和雌激素(E2)水平;计算子宫、卵巢和阴道指数;逆转录PCR法测定下丘脑促性腺激素释放激素(GnRH)、Kiss-1、G蛋白偶联受体54(GPR54)mRNA和垂体GnRH受体(GnRHR)mRNA表达水平。结果 与溶剂对照组比较,模型组大鼠VO,D1和E1分别提前了14.6 d,14.5 d和11.3 d(P<0.05),阴道指数相对升高了151%(P<0.05);模型组大鼠血清中LH,FSH和E2水平明显升高(P<0.05);GnRH,Kiss-1,GPR54和GnRHR mRNA表达量均显著增加(P<0.05)。结论 应用17β-雌二醇可以使雌性大鼠正常的性发育提前,可以考虑用于建立真性性早熟模型。
OBJECTIVE To establish a precocious puberty model in female rats induced by 17β estradiol (E2). METHODS Female Sprague-Dawley rats at 15 d of age were ig given E2 50 pg. kg-1 daily for 5 d. The days of vaginal opening ( VO), first diestrus ( D1 ) and first estrus ( E1 ) in regular estrous cycle were observed. Serum levels of luteinizing hormone ( LH), follicle stimulating hormone (FSH) and estradiol ( E2) were measured by ELISA. The coefficients of ovaries, vaginas, uteruses were calculated, and the mRNA expression of hypothalamic gonadotropin releasing hormone (GnRH), Kiss-1, G protein-coupled receptor 54 (GPR54) mRNA and pituitary GnRH receptor (GnRHR) mRNA was assayed by reverse transcriptase-PCR. RESULTS Compared with vehicle control group, VO, D1 and E1 in model female rats were advanced as much as by 14.6 d, 14.5 d and 11.3 d, respectively (P〈 0.05); the coefficient of vaginas increased by 151% as much as( P 〈 0.05); the serum levels of LH, FSH and E2 increased; and the mRNA expressions of GnRH, Kiss-1, GPR54 and GnRHR significantly increased in model group to 155%, 130%, 900% and 175%, respectively ( P 〈0.05). CONCLUSION A precocious puberty model in female rats can be successfully established by E2.
