摘要
目的:建立高效液相色谱法(HPLC-UVD)测定速溶咖啡中丙烯酰胺的新方法。方法:用水提取速溶咖啡中丙烯酰胺,固相萃取柱进行样品净化后,高效液相色谱法测定。结果:在优化的色谱条件下,丙烯酰胺浓度为0.11μg/ml~100μg/ml时,色谱峰面积与浓度呈良好线性关系,相关系数为0.9999,检出限为0.06μg/ml。咖啡样品加标回收率为97.60%~104.50%,相对标准偏差0.29%~0.50%。结论:该方法具有简便、快速、灵敏度高等特点,适用于测定速溶咖啡中丙烯酰胺。
Objective:A novel method for determination of acrylamide in instant coffee by high performance liquid chromatography was developed.Methods: The acrylamide in the sample was extracted by water,purified with solid phase extraction,then detected by high performance liquid chromatography.Results: Under the optimal chromatography conditions,acrylmide was easily separated from other impurities.The linearity range of the method was 0.11 μg/ml^100 μg/ml with a correlation coefficient of 0.9999.The detection limit of the method was 0.06 μg/ml.The recoveries were 97.60%~104.50% with a RSD of 0.29%~0.50%.Conclusion: The method has advantage of simplicity,fast and sensitivity.And it was suitable for determining acrylamide in the instant coffee.
