摘要
目的:建立高效液相色谱法(HPLC-UVD)测定速溶咖啡中丙烯酰胺的新方法。方法:用水提取速溶咖啡中丙烯酰胺,固相萃取柱进行样品净化后,高效液相色谱法测定。结果:在优化的色谱条件下,丙烯酰胺浓度为0.11μg/ml～100μg/ml时,色谱峰面积与浓度呈良好线性关系,相关系数为0.9999,检出限为0.06μg/ml。咖啡样品加标回收率为97.60%～104.50%,相对标准偏差0.29%～0.50%。结论:该方法具有简便、快速、灵敏度高等特点,适用于测定速溶咖啡中丙烯酰胺。
Objective：A novel method for determination of acrylamide in instant coffee by high performance liquid chromatography was developed.Methods： The acrylamide in the sample was extracted by water,purified with solid phase extraction,then detected by high performance liquid chromatography.Results： Under the optimal chromatography conditions,acrylmide was easily separated from other impurities.The linearity range of the method was 0.11 μg/ml^100 μg/ml with a correlation coefficient of 0.9999.The detection limit of the method was 0.06 μg/ml.The recoveries were 97.60%~104.50% with a RSD of 0.29%~0.50%.Conclusion： The method has advantage of simplicity,fast and sensitivity.And it was suitable for determining acrylamide in the instant coffee.
