摘要
蛋白质-蛋白质相互作用在细胞生命活动中起着十分关键的作用,包括信号转导,细胞骨架的重新构建以及转录控制。重新设计蛋白质-蛋白质相互作用可以指导新药开发,疾病治疗,可以为探讨重大疾病的机制提供重要的理论基础。由于蛋白质之间通过复杂多样的界面发生作用,不同的蛋白质相互作用的界面氨基酸残基组成以及残基之间的相互作用倾向性都有所不同,因此对蛋白质-蛋白质相互作用界面进行研究设计就显得尤为重要。近年来关于蛋白质相互作用界面的设计已有不少成功的例子。该文对蛋白质界面设计的研究进展进行了综述,主要包括蛋白质相互作用界面的性质,结构特点以及它的研究方法等。
Protein-protein interactions plays a crucial role in cell activities, including signal transduction, cytoskeletou rebuild, and transcriptional control. To redesign the protein-protein interaction can guide the development of new drugs and disease treatment. It can also provide important theoretical basis for exploring the mechanisms of major diseases. Because protein-protein interaction occur through a complex and diverse interface, and different protein interfaces have different residues composition and propensity. So it is important to design or redesign protein-protein interface. Recently, many successful examples of protein-protein interface design have been resported. These examples mainly include the follwing two methods. One is redesign of the protein-protein interface and the other is de novo method. This article reviewes the research progress of the protein-protein interface design,including the nature of the protein-protein interaction interface, structural features of interface, as well as some research methods of protein-protein interface.
