摘要
目的:建立酚试剂分光光度法检测亚单位流感疫苗中甲醛含量的方法。方法:采用酚试剂分光光度法,通过改变酚试剂,盐酸,硫酸铁铵浓度,反应时间优化检测条件。结果:在反应体系5 mL中,样本500μL与0.85 mg.mL-1酚试剂2 mL35℃反应10 min后,加入0.1 mol.L-1盐酸2mL和1%硫酸铁铵0.5 mL,35℃反应20 min为最佳反应条件,在0.1~5μg.mL-1范围内呈线性相关,样品加标回收率为98.5%。结论:该方法适用于实验室对亚单位流感疫苗中微量甲醛含量的常规检测。
Objective:To develop a MBTH spectrophotometric method for the determination of formaldehyde in sub-unit influenza vaccine.Methods: The detection condition was optimized by altering the concentration of MBTH,HCl and ammonium ferric sulfate and by changing the reaction time using MBTH spectrophotometric method.Results: In the 5 mL reaction system,the 500 μL sample and 2 mL 0.85 mg·mL^-1 MBTH solution reacted for 10 min in 35 ℃ water bath,and then were added with 2 mL 0.1 mol·L^-1 HCl and 0.5 mL 1% ammonium ferric sulfate,and the best reaction condition was set at 20 min in 35 ℃ water bath.The assay was linear from 0.1μg·mL^-1 to 5 μg·mL^-1.The recovery rate was 98.5%.Conclusion: The method is suitable for the routine detection of trace formaldehyde in sub-unit influenza vaccine.
