Synergistic effect of 5-fluorouracil and cisplatin on tumor cells

摘要 目的观察5-氟尿嘧啶(5-Fu)和顺铂(cP)联合使用对肿瘤细胞是否具有协同抗增殖的作用。方法利用蛋白组织学分析,包括双向凝胶电泳及基质辅助激光解吸/离子化-飞行时间质谱来探索5-Fu或/和CP治疗后与抗增殖相关的蛋白。结果5-Fu处理后有25种蛋白上调节和16种蛋白下调,CP处理后有20种蛋白上调和11种蛋白下调。5-Fu和CP联合使用的最佳浓度为5-Fu:0.5μmol/L和CP:1.0μmol/L,两者联合使用72h后出现协同效应,其协同效应是通过调节膜凋亡受体通路和线粒体凋亡通路而实现的,同时联合使用2种药物后宫颈癌细胞凋亡的敏感性提高。结论5-Fu和CP联合使用可通过协同诱导细胞凋亡而抑制宫颈癌细胞的增殖。 Objective - To investigaie the synergistic effect of 5-fluorouracil (5-Fu) and cisplatin on cervical carcinoma cells in vitro. Methods Using proteomics analyses, including two-dimensional gel electrophoresis and matrix assisted laser desorption/ionization-time of flight mass spectrometry ( MALDI- TOF-MS), we investigated the antiproliferative effect-related proteins after treatment with 5-Fu and/or CP. Results It was found 25 proteins were up-regulated and 16 down-regulated after 5-Fu treatment as deter- mined by spot volume, and 20 proteins were up-regulated and 11 down-regulated after CP treatment as deter- mined by spot volume. The optimal concentrations for combined use of 5-Fu and CP were as follows: 5-Fu: 0. 5 μmol/L, and CP: 1.0 pLmol/L, and the synergistic effects occurred after 72 h. The combination of 5-Fu and CP engaged two apoptotic pathways including the membrane death receptor-mediated apoptosis pathway and the mitochondrial apoptotic pathway. Moreover, the combination of 5-Fu and CP resulted in a remarkably increasing susceptibility to apoptosis. Conclusion The combination of 5-Fu and CP suppresses the growth of cervical carcinoma cells by synergistic effect with the induction of apoptosis. In vitro the synergistic effect of 5-Fu and CP may provide the theoretical guidance for clinical treatment of cervical cancer patients.
