摘要
目的观察复方丹参滴丸对野百合碱所致的大鼠肺动脉高压的影响。方法将健康雄性SD大鼠随机分为5组,正常对照组、模型组、西地那非组、复方丹参滴丸低剂量组、复方丹参滴丸高剂量组。正常对照组一次性腹腔注射生理盐水,其他各组一次性腹腔注射野百合碱60 mg/kg,次日各组给予相应药物,每天1次,持续4周。实验终点,经微导管介入测定大鼠平均肺动脉压,染色观察肺动脉结构的改变,计算肺动脉管壁厚度系数和管壁面积系数。结果野百合碱注射4周后,大鼠出现显著的肺动脉压增高。而大鼠在连续给予4周西地那非(10 mg/kg)和复方丹参滴丸(125、250 mg/kg)灌胃处理后,平均肺动脉压、肺动脉管壁厚度系数和管壁面积系数均较模型组显著性减轻(P<0.05)。结论复方丹参滴丸可有效降低大鼠平均肺动脉压及改善大鼠肺小动脉重构。
AIM To observe the effects of Compound Danshen Dripping Pills on rats with pulmonary hypertension induced by monocrotaline.METHODS Fifty SD rats were randomly divided into 5 groups,normal control group,model group,sildenafil group and low,high doses of Compound Danshen Dripping Pills groups [125 mg/(kg·d) and 250 mg/(kg·d)].The normal control group were injected with saline,and other groups were given a single intraperitoneal injection of monocrotaline 60 mg/kg.On the next day,the low,high dose of Compound Danshen Dripping Pills groups were treated with sildenafil and Compound Danshen Dripping Pills for four weeks.Then mean pulmonary artery pressure of the rats was measured by the micro-catheter.Structural change was observed by staining and the pulmonary artery wall thickness and wall area were calculated.RESULTS Injection of monocrotaline for four weeks caused the increase in pulmonary arterial pressure.Gastric perfusion of sidenafil and Compound Danshen Dripping Pills could decrease the values of pulmonary artery wall thickness and area index as compared with the model group(P0.05).CONCLUSION Compound Danshen Dripping Pills could reduce pulmonary arterial pressure and improve the pulmonary arterial remodeling in rats with pulmonary hypertension.
