复方薤白胶囊对COPD模型大鼠MMP-9和TIMP-1表达的影响 被引量:7

摘要 目的探讨复方薤白胶囊对慢性阻塞性肺疾病(COPD)模型大鼠MMP-9和TIMP-1表达的影响。方法用气管注射脂多糖及熏烟联合造模方法制备COPD大鼠模型,60只大鼠按随机表分为3组:正常对照组、模型组及复方薤白胶囊组。正常对照组、模型组灌胃10mL/kg纯净水,治疗组灌胃给药,连续14d。光镜下观察大鼠病理学改变;用免疫组化法测量支气管-肺组织中MMP-9、TIMP-1蛋白阳性表达面积比率。结果模型组大鼠支气管-肺组织有慢性阻塞性肺疾病的病理改变,复方薤白胶囊组支气管-肺组织的相关损伤和气管壁厚度及平滑肌厚度得到改善;模型组支气管-肺组织中MMP-9、TIMP-1表达较正常对照组升高,差异均具有统计学意义(P<0.05);复方薤白胶囊组MMP-9、TIMP-1表达较模型组明显降低(P<0.05)。结论复方薤白胶囊对COPD大鼠模型的支气管-肺组织的相关损伤起一定保护作用,可减轻炎症反应和支气管壁及平滑肌增厚;有效降低COPD大鼠支气管-肺组织MMP-9、TIMP-1的分布及表达,较好地修复和缓解COPD模型的病理改变,抑制COPD大鼠模型的气道重塑,延缓了COPD的发展。 OBJECTIVE To investigate the effect of CMOC on the expression of MMP-9 and TIMP-1 in rats with COPD. METHODS The rat model of COPD was established by cigarette smoking combined with intratracheal injection of lipopolysac- charide(LPS). 60 rats were randomly divided into three groups: normal control group, model group and CMOC treatment group. The normal group and model group were intragastrically given the 10 mL/kg pure water, the treatment group were giv- en CMOC by stomach, for 14 consecutive days. Pathological changes were observed under light microscopy; positive expres- sions of MMP-9 and TIMP-1 area ratios in the bronchial-lung tissue were determined using immunohistoehemical method. RE- SULTS Characteristic pathological changes of COPD was found in the model group. The bronchial wall and smooth muscle layer thickening in treatment group were significantly decreased. Expression of MMP-9 and TIMP-1 in the model group were higher than normal group, the differences were statistically significant (P〈0.05); compared with model group, the expres- sion of MMP-9 and TIMP-1 of CMOC in treatment group were significantly decreased (P〈0.05). CONCLUSION CMOC can protect bronchial-lung tissue, The expression and distribution of MMP-9, TIMP-1 were reduced, the airway remodeling of COPD rats model was inhibited and the development of COPD was effectively delayed.
