摘要
试验研究一株凝结芽孢杆菌BCRC11592的耐高温、耐酸、耐胆盐及对革兰氏阴性菌抑制的能力,并分析培养时间、温度、初始pH值、接种量和摇床转速等因素对菌体生长和产孢率的影响。结果证实该菌株芽孢有良好的抗逆性,具备开发为微生态饲料制剂的潜力。经优化后其最佳培养条件为培养时间24 h、温度40℃、初始pH值6.5、接种量106cfu、摇床转数140 r/min,在此条件下菌液活菌数为3.5×108cfu/ml,芽孢数为3.0×108cfu/ml,产孢率为85.7%。
This paper studied the stress resistance of the spores of Bacillus coagulans BCRC11592 to heat,acid ,bile salt and bacteriostatic ability against Gram-negative bacteria,and investigated the influence on cell growth and sporulation rate of incubation time,temperature,initial pH value,inoculum and rotation speed.The results confirmend the spores of this strain have good resistance for the potential application in probiotics and the optimal culture conditions were obtained by single-factor lest test as follows :incubation time 24 h ,temperature 40℃ ,initial pH 6.5,inoculum size 10^6 cfu,rotation speed 140r/min.The number of viable cells reached 3.5×10^8 cfu/ml,the number of spores reached 3.0×10^8 cfu/ml,and the sporulation rate was 85.7%.
