芬太尼对MCF-7乳腺癌细胞增殖及迁移和凋亡的影响

摘要 目的:探讨芬太尼对MCF-7乳腺癌细胞增殖、迁移和凋亡的影响。方法:将MCF-7乳腺癌细胞分为6组,正常对照组(C组),芬太尼组(F1组、F2组、F3组、F4组、F5组),芬太尼的终浓度分别为1、0.1、0.01、0.001、0.0001μmol/L,在含去激素新生小牛血清的无酚红培养基培养条件下,分别采用MTT法、划痕实验、Annexin V-FITC细胞凋亡检测试剂盒结合流式细胞仪观察芬太尼对MCF-7乳腺癌细胞增殖、迁移和凋亡的影响。结果:芬太尼组各组OD值、迁移距离和凋亡率与C组比较差异无统计学意义(P>0.05)。结论:在含去激素新生小牛血清的无酚红培养基培养条件下,芬太尼对MCF-7乳腺癌细胞增殖、迁移和凋亡无明显影响。 Objective: To study the effects of fentanyl on the proliferation , migration and apoptosis of human breast carcinoma cell line MCF-7. Method: The MCF-7 ceils were divided into six groups : normal control group ( group C ), fentanyl groups ( group F1, group F2, group F3, group F4, group F5 ), fentanyl groups were exposed to fentanyl 1, 0.1, 0.01, 0.001, 0.0001 μmol/L respectively.MCF-7 cells grown in Phenol red-free media containing fetal bovine serum which had been removed of hormones. The effects of fentanyl on the proliferation , migration and apoptosis of human breast carcinoma cell line MCF-7 were assessed by MTr method, scratch wound assay and Annexin V-FITC apoptosis kit respectively.Result: Compared to the control group, there was no significant difference revealed in the optical density , migration distance and apoptosis rate.Conclusion: When grown in Phenol red-free media containing fetal bovine serum which had been removed of hormones, there is no significant influences of fentanyl on MCF-7 breast cancer cells, proliferation, migration and apoptosis rate.
