干扰素-γ、干扰素-α2b联合应用诱导MCF-7乳腺癌细胞凋亡及机制 被引量:2

摘要 目的研究干扰素-γ、于扰素-α2b联合应用对MCF-7乳腺癌细胞的诱导凋亡作用及其机制。方法流式细胞仪(FCM)检测处理后细胞凋亡及Fas/FasL蛋白表达情况。结果处理后凋亡细胞百分率及FasL的表达增加,随着处理时间的延长,表达有增加趋势,差异有统计学意义(P<0.05);Fas的表达无明显变化。结论干扰素-γ、干扰素-α2b联合应用可诱导MCF-7乳腺癌细胞凋亡,机制可能与上调FasL的表达,加强Fas/FasL之间的交联及使Fas/FasL通路敏感性增加有关。 Objective To investigate apoptosis of MCF -7 treated by interferon and its mechanism. Methods To detecte the apoptosis and Fas/FasL of MCF - 7 treated by interferon through flow cytometry. Results The percentage of apoptotic ceils and expression of FasL increased after treated, and as the extened of time, the percentage of apoptotie ceils and the expression of FasL increased, the difference was considered significant ( P 〈 0.05 ) ; The expression of Fas did not changed. Conclusion Interferon can induce apoptosis in MCF - 7. Mechanism of the apoptosis may be related to upregnlated the expression of FasL and increased the sensitives of Fas/FasL in MCF -7.
