摘要
目的通过整体、细胞、分子三个水平实验,初步研究阿魏药酒的抗肿瘤作用。方法建立小鼠H22肝癌皮下移植模型,灌胃阿魏药酒后测定肿瘤体积、体质量、免疫器官指数和血清中Col-I、CAT、T-SOD活力;采用MTT法测定不同浓度阿魏药酒对小鼠脾淋巴细胞增殖和肿瘤细胞生长的影响;明胶基质金属酶谱法考察阿魏药酒对基质金属蛋白酶表达的影响。结果本实验剂量下,与对照组比较,阿魏药酒灌胃可使肿瘤生长减慢,抑瘤率为12.8%,对各脏器指数均无显著性影响,可使血清中Col-I、T-SOD和CAT活力增强,给药期间不影响小鼠生长状态;实验所用阿魏药酒浓度对小鼠乳腺癌4T1细胞生长无明显影响,低浓度能促进小鼠脾淋巴细胞增殖;阿魏药酒可降低MMP-2和MMP-9的表达。结论阿魏药酒对肿瘤细胞无直接生长抑制作用,可阻止肿瘤的浸润和转移,与具有细胞毒作用的其他中药联合用于抗肿瘤治疗更合理,其抗肿瘤作用可能与其免疫调节和抗氧化作用有关。
Objective To study the anti-tumor activity of Awei medicinal liquor in vivo,at cellular and molecular levels.Methods The anti-tumor activity was tested in vivo by measuring tumor volume,weight,index of immune organs and activity of Col-I,CAT,T-SOD in serum.In vitro the impact of Awei medicinal liquor on tumor cells and spleen lymphocytes was investigated by MTT assay and the matrix metalloproteinase was investigated by Gelatin matrix method.Results Compared with the model group,Awei can suppress tumor growth under the experiment dose with 12.8% inhibitory rate.Awei can enhance the activity of Col-I,T-SOD and CAT in serum although it has no effect on the organ index.Awei at used doses has no effect on 4T1 murine mammary cancer cells and could promote proliferation of murine spleen lymphocytes in low concentrations.Awei is able to inhibit MMP-2 and MMP-9 activity.Conclusion Awei has no inhibition on tumor cells directly but plays a role in resisting tumor metastasis,which suggests that it may be more reasonable to combine with other traditional Chinese medicine with cytotoxicity for cancer treatment.Its anti-tumor effect is associated with immune function and ant-oxidation.
