摘要
目的:观察猪苓和泽泻对肺纤维化大鼠模型血清TGF-β1、TNF-α的影响。方法:将72只健康雄性Wistar大鼠随机分为6组,以支气管内注射博莱霉素的方法制作肺纤维化动物模型。给药后第14天、28天光镜下观察大鼠肺组织病理变化情况,ELISA法检测各组血清中TGF-β1、TNF-α的浓度。结果:各组病理学指标提示大鼠肺组织出现明显的肺间质纤维化,提示动物模型复制成功。通过肺纤维化模型大鼠的病理学改变及血清内TGF-β1、TNF-α浓度比较,各给药组与模型对照组均有显著性差异(P<0.05)。醋酸泼尼松组的指标改善程度最为显著,猪苓加泽泻组次之,猪苓组及泽泻组最弱。结论:猪苓、泽泻及其合用可降低肺纤维化大鼠血清中TGF-β1、TNF-α浓度,减轻博莱霉素诱发的肺纤维化进程。
Objective : To investigate Zhuling, Zexie and Their Combined Use on the concentrations of TGF- β1and TNF-α of pulmonary fibrosis rats and explore the mechanism of treating pulmonary fibrosis with Zhuling, Zexie and Their Combined Use.Methods : Seventy-two male Wistar rats were divided randomly into six groups, Then rats induced pulmonary fibrosis with bleomycin endotracheally.Half rats were sacrificed on the fourteen day and the left ones were sacrificed on the twenty-eighth day.The rat lung histopathology was examined with HE staining and the concentrations of TGF-β1 and TNF- α in the blood serum were examined by Elisa.Results : Changes of pulmonary histopatbology in each group were typical, pointed out that Pulmonary Fibrosis Rats Model were successful.The concentrations of TGF- β1 and TNF- α in blood serum in each treatment group were significantly decreased than those in model control group (P 〈 0.05 ) .Compared comprehensively, prednisone acetate group had the best improvement, then the combination of Zhuling and Zexie group, and Zhuling group and Zexie group worst.Conclusion: The concentrations of TGF- β1 and TNF- α in the blood serum of pulmonary fibrosis rats could be significantly reduced by Zhuling, Zexie and Their Combined Use, which may alleviate the degree of bleomycin induced pulmonary fibrosis in rats.
