蚕丝蛋白对动脉粥样硬化模型大鼠脂质代谢的影响

摘要 目的研究蚕丝蛋白对动脉粥样硬化(atherosclerosis,AS)模型大鼠脂质代谢的影响,为蚕丝蛋白治疗AS提供实验依据。方法选用40只雄性SD大鼠以高脂饲料喂养4周建立早期的动脉粥样硬化(AS)模型,并随机分为五组(每组8只),分别为A组:AS模型组;B组:辛伐他汀组;C组:1g.kg-1.d-1蚕丝蛋白组;D组:2g.kg-1.d-1蚕丝蛋白组;E组:3g.kg-1.d-1蚕丝蛋白组。另选8只以基础饲料喂养4周的健康雄性SD大鼠作为F组(空白对照组)。B组大鼠以10mg.kg-1.d-1辛伐他汀悬浊液灌胃。C、D、E组大鼠以灌胃方式分别给与不同剂量的蚕丝蛋白。A组和F组给予等体积生理盐水灌胃,实验周期为4周。分别于治疗前后检测空腹血脂水平,抗氧化酶含量,并观察血管内皮细胞的结构改变。结果实验结束时,与A组比较,B、C、D和E组的血清总胆固醇(TC),甘油三酯(TG),低密度脂蛋白(LDL-C)明显下降(P<0.05);E组高密度脂蛋白(HDL-C)明显升高(P<0.05);C、D、E组大鼠血清超氧化物歧化酶(SOD)活性明显升高(P<0.05);同时主动脉的管壁结构得到明显改善。结论蚕丝蛋白对高脂饲料诱导的早期动脉粥样硬化模型大鼠脂质代谢紊乱具有明显的改善和调节作用。 Objective To study the effects of fibroin on lipid metabolism in atherosclerosis model rats, so as to provide an experimental evidence for the treatment of fibroin in atherosclerosis patients. Methods 40 male SD rats were chosen and fed with high fat diet for 4 weeks to establish atherosclerosis model. All the rats were divided into 5 groups randomly (8 rats in each group), namely group A (atherosclerosis model group), group B (simvastatin-treated group), group C (1 g. kg-~. dI fibroin-treated group), group D (2 g. kg-1. d-1 fibroin -treated group) and group E (3 g .kg-1 .d-1 fibroin-treated group). Another 8 healthy male SD rats fed with basic diet for 4 weeks were used as group F (blank control group). Rats in group B were treated in the intragastric way with simvastatin turbid liquid at the dose of 10 mg .kg-1. d-1. Rats in C, D and E groups were also treated intragastrically with fibroin at the dose of 1, 2 and 3 g-kg-1 .d-1 respectively for 8 weeks, while rats in A and F groups were treated with the same volume normal saline in the same manner. Fasting serum lipids and antioxidant enzymes were assayed respectively before and after treatment. The microstructure of vascular endothelial ceils were observed at the end of experiment. Results At the end of experiment, compared with group A, total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) of other groups decreased obviously (P 〈0.05), while high-density lipoprotein cholesterol (HDL-C) increased signific antly in group E (P〈0.05). Besides serum superoxide dismutase (SOD) activity increased clearly in all three fibroin-treated groups (P〈0.05). Finally, the microstructural impairment of aorta resulted from high fat diet was improved evidently by all three fibroin-treated groups. Conclusions Fibroin is effective in improving lipid metabolism in atherosclerosis model rats induced by high fat diet.
