摘要 目的应用超声斑点追踪成像技术(STI)观察自发性高血压大鼠左室节段心肌径向(RS)、圆周应变(CS)及应变率的特点,以斑点追踪技术为例,探讨超声应用于鼠科动物模型时的一些注意事项及心得体会。方法 10只Wistar大鼠作为正常对照组,10只SHR大鼠作为阳性组,分别测量两组大鼠左室EF、FS及左室短轴乳头肌平面径向、圆周方向的应变及应变率。结合鼠科动物自身特点、以STI技术用于鼠科动物心脏检查的注意事项等几个方面阐述研究心得。结果两组大鼠的EF、FS测值无明显差异,SHR组RS及RSr较Wistar组显著减低(P<0.05);除了前间隔外,SHR组其余五个节段的CS均低于Wistar组,但组间差异无统计学意义;SHR组六个节段的CSr较Wistar组普遍减低,以前间隔、前壁、侧壁和间隔显著(P<0.05)。两组大鼠径向应变及应变率分布均一,圆周应变及应变率分布不均一。研究鼠科动物模型心脏时首先了解鼠科动物心脏的自身特点,其次做好准备工作,超声检查时注意仪器调节和切面的选择,最后根据研究需要进行心脏结构和功能的相关测定。结论自发性高血压大鼠左室局部心肌的径向、圆周方向的应变及应变率较Wistar大鼠普遍减低。超声在鼠科动物心脏模型中的应用日益广泛,在仪器调节及技术上要多加注意。 Objective To characterize left ventricular myoeardium radial (RS), circumferential strain (CS), strain rate, and speckle tracking among the spontaneously hypertensive rat by ultrasound speckle tracking imaging (STI) , and to share our experience in application of the ultrasound to the murine model. Methods For rat left ventricle, the EF, FS, left ventricular short-axis papillary muscle plane radial, circumferential direction of the strain and strain rate were evalua- ted between normal contrl group (10 Wistar rats) and the positive group (10 SHR rats). Combined with its own characeristics of murine, and research experience to the STI technology for several aspects of the Notes of the murine cardiac examination were introduced. Results The results of two groups were compared. There was no significant difference in EF and FS between these two groups. Results in the SHR group, RS and RSr were significantly:lower ( p d0.05) ; except the interval, CS of the rest five segments were lower than those in the Wistar group, whereas, no significant difference was observed between the two groups; CSr of six segments was generally reduced and was significantly decreased for that of the previous interval, the anterior, lateral walls and the interval ( P 〈0.05). Besides, Radial strain and strain rate distribution was uniform in two groups, but not for circumferential strain and strain rate. During study of murine animal heart models, firstly, we should understand the characteristics of heart in the murine model, secondly, something related was well-prepared, and it was important to make good adjustment of instrument and choice of section ultrasound, and thirdly, the heart structure and function were determined according to our purpose. Conclusion Compared with Wistar rats, left ventricular regional myocardial radial, circumferential direction of the strain and strain rate were generally reduced among spontaneously hypertensive rats.
