摘要
目的探讨砷职业暴露人群Bcl-2基因表达与甲基砷酸的关系,识别砷的遗传毒性.方法整群抽取砒霜厂工人43名作为暴露组,对照组23人.实时荧光定量PCR检测砒霜厂工人外周血淋巴细胞Bcl-2基因,采用氢化物发生原子吸收分光光度法检测尿中各形态砷化合物和总砷含量,并计算一、二级甲基化指数.结果职业暴露工人尿中无机砷、甲基砷酸、二甲基砷酸均显著高于对照人群;职业暴露工人二级甲基化指数显著低于对照人群;职业暴露工人外周血淋巴细胞Bcl-2基因相对表达显著高于对照人群;尿中总砷与Bcl-2相对表达量成显著正相关(r=0.272,P<0.05),MMA与Bcl-2相对表达量成显著正相关(r=0.252,P<0.05),DMA与Bcl-2相对表达量成显著正相关(r=0.280,P<0.05);尿中砷二级甲基化指数与Bcl-2基因相对表达量成显著负相关(r=-0.424,P<0.05).结论砷暴露是工人外周血淋巴细胞Bcl-2基因表达升高的原因,并和甲基化过程密切相关.
Objective To study the relationship between the expression of Bcl-2 gene and monomethylarsonic acid, and study the gentoxicity of arsenic. Methods 43 workers of arsenic factory were selected as exposure group, and 23 people were selected as control group. Bcl-2 gene expression was detected by real-time PCR. We used hydride generation-atomic absorption spectrometry (HGAAS) to determine inorganic arsenic (iAs) , monomethylarsonic acid (MMA) and dimethylarsinie acid (DMA) in urine, and calculated Primary Methylation Index (PMI) and Secondary Methylation Index (SMI). Results The urinary concentrations of iAs, MMA and DMA in the exposed population were significantly higher than in the control group. The SMI in the exposed population was significantly lower than in the control group. The expression level of Bcl-2 gene in the exposed population was significantly higher than in the control group. There was a significant link between the expression of Bel-2 gene and iAs (r=0.272, P〈0.05) , and a significant negative correlation between the expression of Bcl-2 gene and MMA (r=0.252, P〈0.05 ) , and a significant correlation between the expression of Bel-2 gene and DMA (r =0.280, P〈0.05 ) , and a closely negative correlation between the Bcl-2 and SMI (r = -0.424 , P〈0.05 ). Conclusion Arsenic exposure is an obvious reason for the upregulation of Bcl-2 gene and there is a closely correlation between the arsenic methylation products and the expression of the Bcl-2 gene in peripheral blood lymphocytes.
