摘要
为了选择能够适用于悬浮培养生产愈伤组织五味子活性成分的最佳外植体,以北五味子实生苗的幼嫩茎段为外植体,在MS、B5和N6培养基上进行愈伤组织诱导试验,同时采用HPLC法对愈伤组织中的五味子甲素、五味子乙素进行了检测,并对其根、茎段、叶片、愈伤组织和种子等不同组织中五味子甲素和五味子乙素的含量进行了比较分析。结果表明:B5培养基比较适用于愈伤组织的诱导,最适宜的培养基组合为B5+NAA0.2 mg/L+6-BA 1.0 mg/L。切片分析发现,五味子愈伤组织的细胞来源于韧皮纤维以内和木质部以外的这部分细胞,这里即为愈伤组织的起源部位;愈伤组织外形似一朵蘑菇;随着愈伤组织的不断发育,其体积逐渐增大。五味子愈伤组织的形成过程可以划分为诱导起动期、分裂期及形成期这3个时期。高效液相色谱法检测结果表明:五味子甲素和五味子乙素含量最高的是种子。
In order to select out the optimal Schisandra chinensis explants in suspension culture, taking young stems in S. chinensis as explants, callus was induced in MS, B5 and N6. Schizandrin A and B in callus were test by HPLC. The contents of Schizandrin A dnd Schizandrin B in root, stem, leaf, callus and seed were compared. The results show that B5 medium is suitable for callus induction, and the best combination is B5 + NAA 0.2 mg/L + 6-BA 1 mg/L. The results of section analysis indicate that the callus cell in S. chinensis form from the cells within bast fibers and outside of xylem, which is the original site of callus. Appearance of callus likes a mushroom, and its volume is increased gradually with callus development. Formation of callus can be specified into three phases of starting, division and formation. The results of HPLC analysis indicate that contents of Schizandrin A and B in seeds are the highest.
