青藤碱对佐剂性关节炎大鼠行为学特征和滑膜中TNF-α的影响 被引量:12

摘要 目的探讨青藤碱对佐剂性关节炎大鼠关节疼痛和滑膜炎症的影响。方法将40只大鼠随机分为正常组10只和完全弗氏佐剂造模组30只,造模组再分为模型组、青藤碱组和甲氨蝶呤组各10只,青藤碱组和甲氨蝶呤组于造模成功第2天起分别给予青藤碱和甲氨蝶呤干预4周。观察大鼠行为学特征(疼痛阈值),4周后处死大鼠,取膝关节滑膜,行HE染色观察滑膜病理改变,ELISA法检测滑膜中TNF-α的表达。结果模型组疼痛阈值较正常组显著降低,TNF-α较正常组明显增加(P均<0.01);治疗后甲氨蝶呤组和青藤碱组的疼痛阈值和TNF-α表达与模型组比较均有统计学差异(P均<0.05)。结论青藤碱能有效缓解佐剂性关节炎大鼠关节疼痛,减少滑膜炎性因子TNF-α的表达,改善其病理改变,这可能是青藤碱治疗类风湿关节炎的作用机制之一。 Objective To investigate the influence of sinnomenine on joint pain and synovium inflammation in adjuvant-induced arthritis rats. Methods 40 rats were randomly divided into control group ( n = 10) and adjuvant-induced group( n = 30). After being modeled ,the rats in adjuvant-induced group were randomly divided into model group( n = 10), sinnomenine group ( n = 10) and methotrexate group ( n = 10 ). From the second day of modeling success, sinomenine group and methotrexate group were given sinomenine and methotrexat intervention for 4 weeks respectively. The behavioral charac- teristics of rats were observed. HE staining was used to observe synovium pathological changes, ELISA was used to measure the expression of TNF-α in synovium. Results Compared with normal group, pain threshold was significantly lower and TNF-α expressions in synovium were significantly increased in model group ( all P 〈0. 01 ). Compared with the model group, pain threshold and TNF-α expression were statistically different in methotrexate group and sinomenine group( all P 〈0.05). Conclusions SIN could improve the joint pain, decrease TNF-α expression in synovium, and improve the patho-logical changes ; which may be one of its mechanism of action in the treatment of rheumatoid arthritis.
