摘要 目的:观察不同浓度的青藤碱(SIN)体外作用对佐剂性关节炎(AA)大鼠滑膜细胞TNF-αmRNA、IL-1β mRNA、IL-10 mRNA表达的影响。方法:以佐剂性关节炎大鼠为模型,收集滑膜细胞培养,随机分为5组:正常对照组,AA组,AA+SIN 30mg/mL,AA+SIN 60mg/mL,AA+SIN 120mg/mL,反转录PCR检测TNF-α mRNA、IL-1b mRNA、IL-10 mRNA的表达。结果:佐剂性关节炎后滑膜细胞内TNF-αmRNA、IL-1βmRNA、IL-10 mRNA的表达均显著升高,青藤碱在一定的浓度范围内呈浓度依赖性抑制TNF-αmRNA、IL-1b mRNA的表达,而对IL-10 mRNA的抑制效应与浓度无关。结论:青藤碱可能通过降低滑膜细胞内TNF-αmRNA及IL-1βmRNA的表达而发挥治疗AA的效应。 Objective:Effect of different concentration sinomenine(SIN) in vitro on expression of TNF-α mRNA, IL-1βmRNA as well as IL-10 mRNA by synoviocytes in arthritis (AA) rats was observed. Methods:The experimental model of adjuvant AA rats was used. Synoviocytes were collected, cultured and divided into five groups:normal control, AA, AA+SIN 30mg/mL, AA+SIN 60mg/mL, AA+SIN 120mg/mL. TNF-αmRNA, IL-1β mRNA as well as IL-10 mRNA were investigated by the reverse transcription-polymerase chain reaction (RT-PCR). Results:TNF-α mRNA and IL-1β mRNA expression of synoviocytes were markedly increased and inhibited by SIN in a concentration-dependent manner within certain concentration range following adjuvant arthritis. IL-10 mRNA expression was promoted, and inhibition of IL-10 mRNA was independent of SIN concentration. Conclusions:The therapeutical effect of SIN on adjuvant arthritis is probably by decreasing TNF-α mRNA and IL-1β mRNA expression of synoviocytes.
