摘要
目的:研究了正清风痛宁肠溶片在犬体内的药代动力学及绝对生物利用度。方法:采用双周期自身交叉设计,10只Beagle犬分别静脉及口服给予正清风痛宁肠溶片10mg/kg,HPLC法测定血浆中的药物浓度,3p97软件计算二者药动学参数和绝对生物利用度。结果:犬静注给予青藤碱后,药-时曲线呈二室模型,T1/2β为106.67±12.2min,AUC0~t为(93.32±8.07)μg/min·ml。犬口服正清风痛宁肠溶片后,药-时曲线为一室模型,T1/2(Ka)为(36.0±7.97)min,T1/2(Ke)为(84.77±19.33)min,Tmax为(79.27±15.62)min,Cmax为(0.18±0.049)μg/ml,AUC0~t为(38.51±13.23)μg/min·ml。口服青藤碱的绝对生物利用度为41.26%。结论:正清风痛宁肠溶片生物利用度较高,在临床能产生较好的疗效。
