养精胶囊对老年大鼠阴茎海绵体P-Erk1和P-Akt1表达的影响 被引量:7

Regulatory effect of Yangjing Capsule on the expressions of phosphor-Erk1 and phosphor-Akt1 in the corpus cavernosum of aged rats

摘要 目的:通过观察养精胶囊对老年大鼠阴茎海绵体中P-Erk1和P-Akt1表达的影响,探讨其可能的作用机制。方法:将40只24月龄老年雄性SD大鼠随机分为实验组和对照组,每组20只。用药前实验组和对照组各取10只用于eNOS表达测定。实验组(n=10)给予养精胶囊[0.5 g/(kg.d)]灌胃,对照组(n=10)给予等剂量的生理盐水灌胃。4周后,处死所有大鼠,留取大鼠阴茎海绵体组织,HE染色,采用RT-PCR和免疫组化方法检测eNOS、P-Erk1及P-Akt1在阴茎海绵体组织中的表达。结果:HE染色结果显示实验组老年大鼠阴茎海绵体组织血管较对照组明显充盈,管径明显增大。免疫组化结果显示,对照组大鼠阴茎海绵体组织中eNOS的表达干预前(19.35±1.50)和干预后(18.15±1.98)无显著性差异(P>0.05);实验组老年大鼠阴茎海绵体组织中eNOS的表达干预后(24.10±2.40)较干预前(18.80±2.04)显著增强(P<0.05);而干预后老年大鼠阴茎海绵体组织中eNOS的表达,实验组较对照组明显增强(P<0.05);大鼠阴茎海绵体组织中P-Erk1的表达,实验组明显低于对照组(实验组P-Erk1表达量为0.71±0.13,对照组P-Erk1的表达量为0.83±0.17,P<0.01)。而在同等内参GAPDH条件下,两组大鼠间P-Akt1的表达强度未见明显差异(实验组P-Akt1的表达量为0.58±0.17,对照组P-Akt1的表达量为0.48±0.13,P>0.05)。结论:P-Erk1和P-Akt1在老年大鼠阴茎海绵体组织中均有表达,养精胶囊能改善老年大鼠勃起功能,作用机制可能与阴茎海绵体中P-Erk1的低表达相关。 Objective: To observe the effect of Yangfing Capsule on the expressions of P-Erkl and P-Aktl in the corpus cavernosum of aged rats and to explore its action mechanism. Methods: Forty 24-month-old SD rats were equally randomized into an experimental and a control group, 10 selected from each group for detection of the eNOS expression before medication. The rats in the experi- mental group (n = 10) were treated intragastrically with Yangiing Capsule, while those in the control with physiological saline, both at 0.5 g per kg body weight per day for 4 weeks. Then all the rats were killed, and the corpus cavernosum tissues harvested for determination of the expressions of eNOS, P-Erkl and P-Aktl by HE staining, RT-PCR and immunohistochemistry. Results: Obvious distension was observed in the experimental group, but not in the control. The expression of eNOS in the rat corpus cavemosum was signif- icantly higher after treatment than before treatment in the experimental group (24.10 ±2.40 vs 18.80± 2.04, P 〈 0.05 ), but presented no obvious difference in the control ( 18.15 ± 1.98 vs 19.35 ±1.50, P 〉 0.05 ), and it was remarkably higher in the former than in the latter (P 〈 0.05 ). The expression of P-Erkl was markedly lower in the experimental than in the control rats (0.71 ±0.13 vs 0.83 ±0.17, P 〈 0.01 ), but that of P-Aktl exhibited no remarkable difference in the same condition of GAPDH between the two groups (0.58 ± 0.17 vs O. 48 ± 0.13, P 〉 0.01 ). Conclusion: Both PErkl and P-Akt were expressed in the corpus cavernosum of the aged rats. Yanking Capsule could improve the sexual function of aged rats, which might be associated with the lowered expres- sion of P-Erkl.
