摘要
目的研究潜阳育阴颗粒(鬼针草、山茱萸、何首乌、玄参、川牛膝和泽泻)含药血清对血管紧张素Ⅱ(AngⅡ)诱导人脐静脉内皮细胞损伤NADPH氧化酶、蛋白激酶(AKT)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的影响。方法 SD大鼠随机分为正常组、缬沙坦组、潜阳育阴颗粒组,制备含药血清。人脐静脉内皮细胞分为5组,除正常组外,其余组均采用10-6mol/L AngⅡ干预建立人脐静脉内皮细胞损伤模型。造模后,分别给与空白血清培养液、缬沙坦血清培养液、潜阳育阴颗粒血清培养液。TBA法检测MDA水平;ELISA法检测NADPH氧化酶、TNF-α、IL-6水平;Western-Blot检测磷酸化AKT1与AKT1蛋白表达。结果潜阳育阴颗粒显著降低AngⅡ诱导人脐静脉内皮细胞损伤后NADPH氧化酶的活性水平,降低p-AKT1蛋白表达,降低TNF-α、IL-6的释放。结论潜阳育阴颗粒可以抑制AngⅡ诱导人脐静脉内皮细胞损伤后的炎症反应,推测其机制与抑制NADPH氧化酶、AKT、TNF-α、和IL-6有关。
AIM To investigate the effect of Qianyang Yuyin Granules (QYYYG) (Bidens bipinnata Linn, Corni Fructus, Polygoni multiflori Radix, Scrophulariae Radix, Cyathulae Radix and Alismatis Rhizoma ) medica- ted serum on Angiotensin Ⅱ (AngII) -induced NADPH, AKT, tumor necrosis factor-alpha (TNF-α) and inter- leukin-6 (IL-6) in human umbilical vein endothelial cells (HUVECs). METHODS Rats were randomly divided into three groups receiving Valsartan, QYYYG, and normal food, respectively. Drug-containing serum was pre- pared. Except for normal group, HUVECs were incubated with 10-6 mol/L AngII to form the damaged HUVEC models. These HUVECs were cultured with prepared serum containing Valsartan, QYYYG, and blank, respective- ly. The intervention was observed. NADPH, TNF-α, IL-6 were determined by enzyme linked immune-sorbent as- say and the content of MDA was measured by the method of thibabituric acid. Western-blot was used to detect the protein expression of phosph-AKT1 and AKT1. RESULTS QYYYG could lower the level of NADPH activity , the expression of p-AKT1, and the release of TNF-α and IL-6 in the damage to HUVECs induced by 10-6mol/L Angiotensin Ⅱ. CONCLUSION These results indicate that QYYYG can significantly inhibit the inflammation of HUVEC induced by AnglI related to NADPH, AKT, TNF-α and IL-6.
