谷氨酸生产菌T6-13的噬菌体的血清学和DNA内切酶分析 被引量:1

摘要 从13个省市的39家工厂,采集73份土和水标样。从中分离到63份噬菌体,阳性率占86.3%。电镜观察20株噬菌体,形态均属Akermann分类的B1型,即有一直径略有差别的正六角形的头和较长的非收缩弯曲的尾部。所分离的噬菌体均能裂解谷氨酸生产菌T6-13。形成的噬菌斑有4种类型。根据噬菌体形态、噬菌斑类型、对抗性菌株的裂解能力,选择一批制备抗血清,借交叉中和试验将63株噬菌体分成12个血清型。检查12个血清型噬菌体对不同的抗噬菌体突变株的裂解谱,有4种类型菌株,其中4-210菌株为具有抗噬菌体谱广的特性。结合对不同抗性菌株裂解能力的差异,12个血清型噬菌体又可分成15种血清—裂解类群。取其中21株噬菌体DNA用BamHⅠ酶切,出现16种不同电泳谱型。 Glutamate-producing bacterium T6-13 strain was extensively used in our country to produce glutamic acid presently.Its phages were widespread in the environment in these factories. Phage contamination is extremely harmful in the production of glutamic acid. Sixty three strains of ph-age were isolated from 73 soil and sewage samples collected from 39 gluta-mateproducing factories. 20 examined of them belongs to Akermann Bl type under electron microscope.Four types of phage plaque were formed on the glutamate-producing bacterium. By using anti-phage sera prepared from 7 phages, the 63 phages were divided into 12 serotypes by neutralization test. The original T6-13 and its 3 mutant strains of baderia were examined for susceptibility and resistance to the 12 serotypes of phages. The mutant strain 4-210 was found to be resistant to all 12 serotypes. When the phage DNAs were digested with endonuclease BamHI, 16 kinds of electroph-oresis pattern were observed.
