摘要
本文比较了海藻酸钙、明胶-戊二醛、聚丙烯酰胺和琼脂包埋法制备固定化细胞的方法。前者较后几种有较大的优越性,是一种较为理想的固定化方法.分析了海藻酸钙法国定化细胞各主要因素对酶活性的影响,确定出制备固定化细胞的最佳条件.观察凝胶珠中细胞及酶活性的变化表明,细胞在凝胶珠中生长良好,有利于谷氨酸生产.固定化细胞发酵谷氨酸的最适 pH 为7. 0~7. 5,最适温度32℃,最适转速140rpm.每批发酵96小时,糖利用率为98%,产酸量达20g/L,比常规游离细胞增加15%.
Four different methods of immobilizing corynebacterium glutamicum whole cell systems were investigated.There are several advantages of the cell entrapped in calcuim alginate gel lattice over those entrapped in a polyacrylamide,gelatin,and ager gellattice,such as higher enzyme activity,enough mechanical strength and the process was simple,Immobilized cells are not only living,but also growing and reproducing well in the gel beads.It is one of ideal immobilizated methods.Several main factors of effect in immobilized cells enzyme activity were also studied simultaneously,the optium conditions for immobilization cell beads with ealeuim alginate were determined.Immobilized cell beads were aerobic cultured in seed culture medium,the grow and changes of cell colony and enzyme activity in bcads were investigated.It was found that cells grow well and the amount of cells and enzyme activity in beads are in relativition to the culture time befor 120h,they are constant after 120h.This is very useful for fermentation of glutamate.The fermentation tests were carried out in 500ml flask,there are 5g immobilized cells and 100ml fermentation medium in it.The optium conditions of fermentation in immobilized cells systems were temperture 32~37℃,pH 7. 0~7. 5,stir speed 140rpm.The yield of immobilized cell glutamate is 20g/L,It is higher 15 percent than that of native cells.
