慢病毒介导TGF-β3、CTGF和TIMP1联合转染对兔退变椎间盘影响 被引量:3

摘要 目的研究慢病毒介导的转化生长因子β3(TGF-β3)、结缔组织生长因子(CTGF)和基质金属蛋白酶组织抑制因子l(TIMP1)联合转染对兔退变椎间盘的影响。方法新西兰大白兔15只,纤维环穿刺法建立椎间盘退变模型,实验组注入携带TGF-β3、CTGF和TIMP1基因的慢病毒颗粒50μL,对照组注入等量空病毒,各组在相同条件下独立培养,分别于术后16、20周对退变椎间盘进行影像学观察。结果实验组与对照组及穿刺组相比,椎间盘退变程度明显减轻,差异有显著性(χ2=9.11、19.05,P<0.05)。结论 TGF-β3、CTGF和TIMPI多基因联合转染后可以起到延缓椎间盘退变的作用。 Objective To study the effect of lentivirus-mediated TGF1β, CTGF and TIMP1 co-transfection on degenera- tive intervertehral disc (DID) in rabbits. Methods A model of DID was created by an anulus needle puncture in 15 New Zealand white rabbits, 50 μL of Lenti-TGF β3-P2A-CTGF-T2A-TIMP1 were injected into DID of rabbits in experimental group, and lenti- virus was injected into DID in those in control group. They were independently cultured under the same condition. Imaging obser- vation of DID was conducted 16 and 20 weeks after the procedures. Results The DID in the rabbits in the experimental group was dramatically relieved as compared with the control and puncture groups, the difference being significant between them (X2 = 9.11,19.05 ;P〈0.05). Conclusion Combined Lenti-TGF β3-P2A-CTGF-T2A-TIMP1 can successfully delay the degeneration of intervertebral discs.
