摘要 目的:通过研究转化生长因子β3(transforming growth factor-β3,TGF-β3)对椎间盘退变大鼠的软骨终板凋亡的影响,探讨TGF-β3在椎间盘退变性疾病中的作用,探索治疗椎间盘退变性疾病的新思路。方法:取87只新生SD大鼠,采用双后肢直立法建立大鼠椎间盘退变模型。饲养18个月后,处死5只SD大鼠,行HE染色,光镜下见椎间盘髓核和纤维环退变,证实造模成功。随机选取存活大鼠中的60只随机分成对照组、0.9%氯化钠液组及TGF-β3组,每组20只。对照组不作任何处理;0.9%氯化钠液组及TGF-β3组分别于腰3、4节段间软骨终板处注射0.9%氯化钠液和TGF-β3,每周1次,共3次。3周后处死所有大鼠,取腰3、4节段椎间盘制成切片,行Tunel染色,计算软骨终板细胞凋亡指数,并进行统计学分析。结果:双后肢直立法成功建立大鼠椎间盘退变模型;0.9%氯化钠液组的凋亡指数较对照组明显升高,差异有统计学意义(P<0.05);TGF-β3组的软骨终板细胞凋亡指数低于0.9%氯化钠组(P<0.05)而高于对照组(P<0.05)。结论:TGF-β3可以抑制椎间盘退变大鼠的软骨终板细胞的凋亡,具有治疗椎间盘退变性疾病的潜在价值。 Objective:To investigate the therapeutic effect of transforming growth factor-β3 (TGF-β3) on intervertebral disc degeneration (IVDD) rat models. Methods:Eight-seven Sprague Dawley (SD) rats were included in this study. The IVDD SD rat model was constructed by a double-hind legs erection method. After eighteen months of feeding, five SD rats were killed and their intervertebral discs were stained with HE. Successful construction of an IVDD model was demonstrated by the nucleus pulposus and annulus fibrosus showing regression under a conventional light microscope. Sixty survived rats were divided into the control group, 0.9% sodium chloride solution group and TGF-β3 group, with 20 rats in each group. The rats in the con- trol group received no treatment. 0.9% sodium chloride solution or TGF-β3 was injected in the cartilage endplates at the second or third lumber vertebration of rats in the 0.9% sodium chloride solution group and TGF-β3 group, respectively, once a week for a total of three doses. After three weeks, all rats were killed and the second or third lumbar spine slices were excised. Then the lumbar spine was stained by Tunel staining. The apoptotic index was calculated. Results: The IVDD model was successfully constructed. The apoptotic index in the 0.9% sodium chloride solution group was significantly higher (P〈0.05) than that in the control group; the apoptotie index in the TGF-β3 group was significantly lower than the 0. 9% sodium chloride solution group (P〈0.05), but significantly higher than that in the control group (P〈0.05). Conclusions: TGF-β3 can inhibit the apoptosis of the intervertebral disc cells in rats and may serve for the treatment of IVDD.
