摘要
目的观察磷酸二酯酶4(phosphodiesterase,PDE4)抑制剂咯利普兰对佐剂关节炎(adjuvant arthritis,AA)大鼠肺功能及Th1/Th2平衡的影响。方法通过注射完全弗氏佐剂(complete freund's adjuvant,CFA)建立AA模型,并给予咯利普兰干预,以小动物肺功能仪测定大鼠肺功能,酶联免疫吸附测定(enzyme-linked immunosorbnent assay,ELISA)法测定血清中肿瘤坏死因子α(tumor necrosis factor-alpha,TNF-α)、白细胞介素10(interleukin-10,IL-10)水平。结果 AA大鼠存在通气功能及小气道功能障碍,咯利普兰能改善肺功能指标(P<0.01);AA大鼠血清中TNF-α显著升高,IL-10显著降低,Th1/Th2增高(P<0.01);咯利普兰使AA大鼠血清中TNF-α减低、IL-10升高、Th1/Th2减低(P<0.01)。结论咯利普兰能下调AA大鼠致炎因子、上调抗炎因子,调节Th1/Th2平衡并改善受损的肺功能,提示PDE4参与了类风湿肺的发病,PDE4抑制剂具有调节免疫治疗类风湿肺的潜能。
Objective To observe the effects of rolipram on lung ruction and Thl/Th2 balance of rats with adjuvant arthritis (AA). Methods The AA model was established by injection of eomplete Freund. adjuvant (CFA) to the rats, and rolipram was given as an intervention, rat lung function was measured by a small animal spirometer, and serum tumor necrosis faetor-α (TNF-α), interleukin-10 (IL- 10) levels were determined by enzyme-linked immunosorbnent assay (ELISA). Results AA rats showed ventilatory dysfunction and small airway dysfunction, rolipram ean improve lung function indicators (P 〈 0.01 ). The serum levels of TNF-α were significantly increased, IL-10 significantly reduced, Thl/Th2 increased(P 〈0.01 ) in AA rats,rolipram reduced the level of TNF-α,increased the level of IL-10, and lowered the Thl/Th2 value (P 〈 0.01 ). Conclusion The results suggested that rolipram could reduce the proinflammatory factor, raise anti-inflammatory factor, regulate Thl/Th2 balance and improve lung function impairment in AA rats. PDE4 may be involved in the pathogenesis of rheumatoid lung, PDE4 inhibitoitors may modulate the immune and has a potential to treat rheumatoid lung in the future.
