摘要
目的:探讨姜黄素对抗环磷酰胺致小鼠精子DNA损伤的保护作用。方法:成年昆明小鼠随机分成4组:正常组,模型组,姜黄素低、高治疗组。模型组和姜黄素低、高组小鼠按照40mg/kg,腹腔注射环磷酰胺(cp),连续5d,同时姜黄素低、高组小鼠每天给予姜黄素混悬液100mg/kg,200mg/kg灌胃;正常组及模型组小鼠予等量0.5%羧甲基纤维素钠灌胃,共28d。观察小鼠精子低渗膨胀率,小鼠睾丸组织匀浆后测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、活性氧(ROS)含量及小鼠精子DNA的完整性。结果:与模型组相比,姜黄素组睾丸组织SOD与GSH-Px含量等均明显升高(P<0.05、P<0.01),MDA、ROS含量明显降低(P<0.05、P<0.01),吖啶橙荧光染色法(AOT)显示:与模型组相比,姜黄素组小鼠精子DNA的完整性明显升高,有明显的统计学意义(P<0.01)。结论:环磷酰胺可破坏小鼠精子DNA的完整性,导致精子核成熟率下降;姜黄素可以改善环磷酰胺对小鼠精子DNA的氧化作用,提高精子DNA的完整性。
Objectives: To explore the protective effect of curcumin against cyclophosphamide -induced sperm DNA damage in mice. Method: We divided adult Kunming mice into four groups randomly. Those are normal group, model group, low cureumin treatment group, high curcumin treatment group. As for the model group, low curcumin treatment group and high curcumin treatment group, we performed 40mg/kg intraperitoneal injecton of cyclophosphamide (ep) for five days. At the same time, we performed 100mg/kg curcumin suspension gavage to low curcumin treatment group and 200mg/kg to high curcumin treatment group every day. As for the normal group and model group, we performed the same amount of 0.5% sodium earboxymethyl cellu- lose gavage. The process lasted for 28 days. Then we observed the sperm hypotonic swelling rate. We detected superoxide dis- mutase (SOD) , glutathione peroxidase ( GSH - Px) , malondialdehyde ( MDA ) , reactive oxygen species ( ROS ) content and mouse sperm DNA integrity. Result : Compared with the model group, the curcumin groups have significantly higher testieular tissue SOD and GSH - Px content ( P 〈 0. 05, P 〈 0.01 ), have significantly lower MDS and ROS (P 〈 0. 05, P 〈 0.01 ). Acridine orange staining (AOT) showed that compared with the model group, the mouse sperm DNA integrity of the eurcumin groups increased obviously and the increase was statistically significant ( P 〈 0.01 ). Conclusion : Cyclophosphamide can damage the mouse sperm DNA integrity which leads the decreased sperm nuclear maturity rate. Curcumin can improve the oxidation of cyclophosphamide in mice sperm DNA, and improve sperm DNA integrity.
