摘要
目的 研究组织因子 (TF)、凝血酶受体 (TR)和血栓调节蛋白 (TM )在胰腺癌细胞中的表达。方法 采用细胞培养、免疫细胞化学和Northern印迹分析方法。结果 每个抗原都在细胞膜及 (或 )细胞浆中有特征性的免疫反应 ,TF和TR主要在细胞浆 ,TM在细胞膜。TR表达越多 ,TM表达越少。TRmRNA在野生型胰腺癌细胞株中表达 ,凝血酶和水蛭素均降低TRmRNA的表达量。凝血酶可致人胰腺癌细胞株MiaPaCa 2和Panc 1DNA合成增加 (P <0 .0 5 ) ,在凝血酶刺激 0 .5和 6h时达高峰。结论 胰腺癌细胞的TF、TR和TM表达参与了胰腺癌的血液高凝状态。
Objective\ To study the expression of tissue factor (TF), thrombin receptor (TR) and thrombomodulin (TM) in human pancreatic cancer cells. Methods\ Human pancreatic carcinoma cell lines ( n =4) were studied by using cell culture, immunocytochemistry and Northern blotting analysis. Results\ Both TF and TR were predominantly cytoplasmic in distribution, TM was membrane localized. The higher TR expression, the lower TM expression. TRmRNA was expressed in 4 pancreatic cancer cell lines (CaPan 2, MiaPaCa 2, Panc 1 and BxPC 3). Thrombin and hirudin decreased the TRmRNA expression. Thrombin treatment increased DNA synthesis of MiaPaCa 2 and Panc 1 cells. Conclusion\ The expression of TF, TR and TM might take part in the pathogenesis of hypercoagulable state in pancreatic cancer.
