摘要
目的:基于代谢组学的方法研究天麻-川芎药对对大鼠尿液中内源性代谢产物的影响。方法:将药材提取物分为正常对照组、天麻组、川芎组、天麻川芎组A(天麻:川芎比为1:4)、天麻川芎组B(天麻:川芎比为4:1)等5组,每组中药材均用10倍量70%的乙醇提取,用HPLC法测定提取物中天麻苷和川芎嗪的含量来确定给药剂量。每组按11.67 g·kg^(-1)的剂量对大鼠连续给药,收集尿样,通过液质联用方法对尿液中内源性代谢物进行全扫描分析,采用MATLAB和SIMCA-P两种数据处理软件对大鼠尿液内源性代谢产物进行主成分分析(PCA)。结果:连续灌胃29 d后,各给药组与正常对照组相比,大鼠尿液内源性代谢产物均发生显著变化。结论:天麻川芎提取物显著影响大鼠尿样中与药理作用相关内源性物质的代谢,为深入研究天麻川芎药对提供依据。
Objective: To study the effect of drug pair of Gastrodia elata and Chuanxiong Rhizoma on the endogenous metabolites in rat urine based on metabolomics. Method : The rats were randomly divided into five groups : the normal control group, Gastrodia elata group, Chuanxiong Rhizoma group, Gastrodia elata and Chuanxiong Rhizoma group A with the compatibility proportion of 1 : 4, Gastro- dia elata and Chuanxiong Rhizoma group B with the compatibility proportion of 4: l. The herbs were extracted by 70% ethanol ( 10 times volume), and HPLC was used to assay the content of gastrodin and tetramethylpyrazine in the extracts to confirm the best admin- istration dose. The intragastric administration dose was 11.67 g kg-1 per day and urine was collected. The endogenous metabolites in rat urine were under full scan analysis by LC-MS, and two data processing softwares ( MATLAB and SIMCA-P) were used in principle component analysis (PCA) of the endogenous metabolites. Result: Compared with that in the normal control group, the endogenous metabolites in rat urine showed significant changes in the other groups after the 29-day administration. Conclusion: Endogenous me- tabolites in rat mine is obviously influenced by Gastrodia elata and Chuanxiong Rhizoma, which, provides the basis for the further study on Gastrodia elata and Chuanxiong Rhizoma drug pair.
