摘要
目的探讨静脉血栓溶解过程中血小板衍生生长因子CC(PDGF-CC)的表达情况及其意义。方法取健康成年雄性SD大鼠建立下腔静脉血栓模型,设对照组及血栓组,血栓组分别于术后1、3、7、14、21、28d收集下腔静脉血管壁及血栓标本(n=12)。应用Western blotting及实时荧光定量PCR分别检测血栓自然溶解过程中大鼠下腔静脉管壁及血栓内PDGF-CC蛋白及mRNA的表达;取术后7d大鼠下腔静脉血管壁及血栓标本(n=3),用于病理组织形态学及PDGF-CC免疫组化观察。结果 Western blotting检测显示,术后1~28d大鼠下腔静脉血栓中均有PDGF-CC蛋白表达,以术后7d表达量最高(P<0.05);Real-time qPCR检测显示,术后1~28d大鼠下腔静脉血栓中均有PDGF-CC mRNA表达(P<0.05),其中术后7d相对表达量最高,为对照组的77.71倍;下腔静脉血管壁中同样有PDGF-CC蛋白及mRNA表达,但表达量均较低,组间比较差异无统计学意义(P>0.05)。免疫组化染色显示术后7d的血栓标本中,新生血管周围有PDGF-CC特异性表达。结论 PDGF-CC在大鼠下腔静脉血栓自然溶解过程中呈较高水平表达,且在血栓中的新生血管周围有特异性表达,提示其在血栓溶解过程中可能具有重要作用。
Objective To explore the expression of platelet-derived growth factor CC (PDGF-CC) during intravenous thrombolysis in rats and the significance thereof. Methods The inferior vena cava thrombosis model was established in adult male SD rat, and control and experimental groups were set up. Specimens of inferior vena cava and thrombus therein (n^12) were collected on day 1, 3, 7, 14, 21 and 28 after modelling. The expression of PDGF-CC protein and mRNA in inferior vena cava and thrombus during thrombolysis was determined by Western blotting and real-time qPCR. The specimens of inferior vena cava and thrombi (n=3) of rats 7 days after modelling were collected to observe the pathohistological and immunohistochemical changes. Results Western blotting and real-time qPCR showed that PDGF-CC protein and mRNA were expressed in inferior vena cava thrombi from 1 to 28 days after modelling, and the peak value of expression was found on day 7(P〈0.05). PDGF-CC protein and mRNA were also expressed in the wall of inferior vena cava, but the expression level was low and no significant difference was found between groups (P〉0.05). Immunohistochemical analysis revealed that the specific expression of PDGF-CC was present surrounding the new vessels in thrombi 7 days after modelling. Conclusions PDGF-CC has been found to express in high level during intravenous thrombolysis in rats, and it specifically expresses around the new vessels in thrombi, implying that PDGF-CC may play an important role during intravenous thrombolysis.
