摘要
马铃薯再生体系的建立是进行马铃薯遗传转化的前提条件。本研究以马铃薯克新1号为植物材料,对马铃薯愈伤组织的诱导、不定芽的分化进行了最佳培养基优化。结果表明,马铃薯诱导愈伤组织的最佳培养基激素浓度是2.5mg/L 6-BA、0.2mg/L NAA,且茎段的诱导率要高于叶片的诱导率。不定芽分化的最佳培养基激素浓度为2.0mg/L 6-BA、0.1mg/L NAA、5.0 mg/L GA3,诱导率为30%,诱导的不定芽在0.2 mg/L NAA生根培养基中可以正常生根。本研究初步建立了马铃薯再生体系,为后续的金黄色葡萄球菌奶牛乳腺炎转基因马铃薯疫苗的制备奠定了基础。
Establishment of regeneration system is a prerequisite of genetic transformation of potato. In this study, the mediums for callus induction and shoot regeneration of potato were optimized with potato Kexin 1 as material. The results showed that the best hormone concentration for callus induction of potato was 2.5rag / L 6 - BA and O. 2 mg/ L NAA, and the induction rate of the stern section was higher than the leaves. The best hormone concentration for shoot regeneration of potato was 2.0rag / L 6 - BA, 0. ling / L NAA, and 5.0 mg / L of GA3, and the induction rate was about 30%, and furthermore, the shoots could root normally in the 0.2 mg / L NAA MS medium. In this study, the regeneration system of potato was established preliminarily, which laid the foundation for the preparation of transgenic potato vaccine against Staphylococcus aureus mastitis.
