摘要
目的研究黏附因子丛生因子A(clumping factor A,ClfA)在金黄色葡萄球菌(SAU)不同菌株中的分布情况,为开发具有广泛保护效果的SAU疫苗奠定基础。方法采用PCR法扩增目的基因,构建重组质粒,以大肠杆菌BL21(DE3)为表达宿主,对目的蛋白进行表达、纯化。PCR及点杂交法检测ClfA在不同菌株中的分布情况。结果得到纯度较高的目的蛋白,PCR及点杂交结果显示ClfA存在于菌株USA300,而在菌株546、1884、1885中并无分布。结论 ClfA在SAU各菌株中分布不广泛,可能难以作为具有广泛保护效果的SAU疫苗组分。
Objective To test the distribution of clumping factor A(ClfA)in different strains of Staphylococcus aureus(SAU).Methods The gene of interest was amplified by PCR and subcloned into the pET21a(+)vector.The recombinant plasmid was transformed into E.coli BL21(DE3)cells to express the target proteins,which were purified by passing the Histrap and the Q Sepharose F.F.column.The distribution of ClfA in different SAU strains was tested by PCR and dot blot.Results Highly purified protein was obtained.There was ClfA in S.aureus USA300,but not in S.aureus 546 or 1884.Conclusion The ClfA is not distributed widely,and may not be a broadly protective vaccine.
